Plant male sterile is a common phenomenon and the basic of using hetorosis to produce seeds,which could make farmer emasculate and reduce the cost. At present,tens of different cytoplasmic-nuclear male sterile lines with stable male sterility have been developed in soybean,great production of soybean has been achieved by the utilization of heterosis.Few studies have been done on the mechanism of the cytoplasmic-nuclear male-sterile lines.In this paper,studies on sixty-two pairs soybean cytoplasmic male sterile lines and corresponding maintainer lines were carry out,their mitochondrial genomic DNA was marked and analysised using random amplified of polymorphic DNA(RAPD),it can lay foundation for the identification of soybean sterile line and maintainer line and mechanism of the cytoplasmic-nuclear male-sterile lines.Part of the specific products we got in soybean cytoplasmic male sterile lines and corresponding maintainer lines were sequenced .Results:1. 5 sucrose gradient (30%,35%,40%,45%,50%)and 2 ultracentrifugation (35000rpm) were set up to ensure the Purity of mitochondria DNA we extract.2. Specific sequences were amplified in maintainer lines by 11 primers:OPA-03 OPD-20 OPE-01 OPG-17 OPH-18 OPK-07 OPM-06 OPP-15 OPQ-12 OPQ-13 OPR-03, specific sequences were amplified in sterile lines by 3 primers:OPD-02 OPL-13 OPN-03,and despite the different sizes, specific sequences were both amplified in maintainer lines and sterile lines by OPB-06 OPI-09 OPL-10.the result may be corresponding to soybean cytoplasmic male sterile.3. Partial of the specific products(8)were sequenced,7 of the 8 specific amplified products have high homology with plant mitochondria genome, except the specific products which were amplified by OPN-03.the result means that the mitochondria DNA we extract have high purity.4. 71bp of the specific sequence amplified by OPA-03 can match the sequence near the upstream of cox1 ORF in Arabidopsis thaliana mitochondrial genome,and share the homology of 98%.So we conclude that soybean cytoplasmic male sterile maybe relate with cox1.
