The established view in cellular biology dictates that the cellular internalization of hydrophilic macromolecules can only be achieved through the classical endocytosis pathway. However, in the past ten year s several peptides have been demonstrated to translocate across the biological membrane by a recepter-independent pathway, these peptides have been used successfully for the intracellular delivery of biologically active protein, compounds and DNA and offers several advantages over conventional techniques.Objective1.To study the structure and function of the membrane penetrating peptides and found new peptide vectors.2.To observe the intercellular transport property of a novel synthesized peptide.3. To further understand its mechanism of internalization.Methods and Results1.A new peptide sequence was designed based on the analysis of the molecular composed of the membrane penetrating peptides using the peptool life software.2.Immunofluorescence technique was used to observe the penetrati on ability and the delivery property of the novel synthesized peptide. 3.The study revealed that internalization occurs within 2 minutes, can be detected at concentration as low as 0.01μmol/L and is not altered by lowering the temperature to 0℃.5.we have further observed that penetration of the synthesized pe-ptide is not blocked even under the condition of pre-fixing cells.Conclusion1.The result implies that the synthesized peptide cross cellular m-embrane using protein-independent mechanism.2.It also suggests that the synthesized peptide can be applied to be a special biological macromolecule vector.
