Recombinant DNA technology is very important in genetic engineering. In this paper,we have constructed two recombinant vectors by it.Cut off betA fragment from plasmid pROK.II with Hindlll and EcoR I as insert,and cut pA into linear plasmid as vector fragment. Link the insert and vector fragment together with T4 ligase,and the new vector with gene betA and gus was constructed.Cut pGAH with Hindlll into linear plasmid as vector fragment and insert betA fragment into it. The recombinant vector with dualt salt-tolerance gene betA and codA was constructed.With haploid as transgenic material can decrease mosaic and can avoid properties separation in transforming. The pollen of this hybrid variety is used as explants and the medium is added some plant hormones,such as KT and 2,4-D in different concentration as experiment component. The optimal medium of callus is MS+24mg/L 2,4-D+2mg/L KT. The best medium of multiplication is MH+BA 0.1-0.5 mg/L+NAA 0.02-0.05 mg/L. The best medium of rooting is l/2MS+0.2mg/L NAA. This system is suit to transformation.
