Studies On The Therapetic Basis Of Chinese Compound Formula Xiao Xian Xiong Soup

Xiao-xian-xiong Soup (XS, is one Chinese compound formula, which is originated from<Shang Han Lun>, written by a famous people named Zhang Zhong Jing, used to treat pneumonia, pleurisy, coronary heart disease, etc. XS is composed of three kinds of crude herbs, Trichosanthes kirilowii Maxim., Coptis chinensis Franch. and Pinellia ternata (Thunb.)Breit. In this paper we adopt phytochemistry, analytical chemistry, synthesis chemistry,pharmacology and pharmacokinetics methods to study on the chemical structure and biological activity of active ingredients in XS; illustrate the therapeutic basis of XS, provide solid scientific basis for the development of new drug and establishment of quality control methods, provide a new idea and method for the compatibility mechanism and therapeutic basis of Chinese compound formula and promote the modernization of Traditional Chinese medicine.The chemical constituents of Trichosanthes kirilowii Maxim. and Coptis chinensis Franch. were studied by kinds of chromatographical techniques (such as silica gel chromatography, sephadex LH-20, PTLC and HPLC). After systematically studied, 34 compounds were purified. Utilizing chemical and spectroscopic methods (UV, NMR, MS),the structures of 32 compounds were fully characterized. From Trichosanthes kirilowii Maxim. there were 19 alkaloids and 3 flavonoids; from Coptis chinensis Franch. 7 alkaloids and 2 lignans were isolated, which were all known compounds.The quality control methods of alkaloids compounds were studies. An HPLC-QTOF-MS method was established to determine the content of alkaloids in Coptis chinensis Franch.through 12 different decocting methods. The assay methods were simple, rapid and with good reproducibility,to provide a quantitative basis for the quality assessment for herb and Xiao-xian-xiong Soup.We adopted a simple synthesis method to produce a series of fatty acid amides compounds such as auroyldiethanolamide and capryldiethanolamide for the purpose of pharmacology activity and pharmacokinetic study.In this paper we adopted pharmacology method, setting up the promting heparin anticoagulant activity in vitro mode, tracking the effective components, the results reveal the fatty acid amides compounds have the significant promoting heparin anticoagulant activity.The developed HPLC-MS method was applied in the pharmacokinetic studies of berberine and palmatine after oral administration of monomeric compound, herbs and Xiao-xian-xiong Soup formulation. The pharmacokinetic characteristics were significantly different between rats with administration of the different groups, because of the effects of co-existing compounds even with the same dosages.
