Over the years, a number of RNAs that do not function as messenger RNAs (mRNAs), transfer RNAs (tRNAs), or ribosomal RNAs (rRNAs) have been discovered. They are noncoding RNA(ncRNA). There are many kinds of ncRNA, such as: mRNA-like RNA, microRNA(miRNA), snRNA, snoRNA, scRNA, pRNA, SRP-RNA, gRNA et al. The functions described for ncRNAs thus far are extremely varied. Some ncRNAs affect transcription and chromosome structure. Some ncRNAs play roles in RNA processing and modification. Some ncRNAs also regulate mRNA stability and translation. Other ncRNAs affect protein stability and transport. ncRNA can act with several mechanisms: 1) Direct base-pairing with target RNA or DNA molecules is central to the function of some ncRNAs; 2) Some ncRNAs mimic the structure of other nucleic acids; 3) ncRNAs also can function as an integral part of a larger RNA-protein complex. mRNA-like RNA and miRNA are two important kinds of ncRNA. They have relationship with cell proliferation and differentiation, embryo development, tumor formation and inhibition.MicroRNAs (miRNAs) are short 20-25 nucleotides RNA molecules that have been shown to regulate gene expressions in a variety of eukaryotic systems. miRNAs are widespread in eukaryotes and several hundred of miRNAs have been identified, but still a lot of miRNAs have not been detected in various eukaryotic organisms. However, it is not an easy work to clone miRNAs by traditional methods. Liver is one of the largest and most important organs in our body, which can build and store many nutrients that our body needs, break down harmful substances, remove waste products from our blood. To study functions of miRNAs in liver, we identified 27miRNAs fetal liver by a new miRNA cloning method and analyzed the effect of some miRNAs on cell proliferation.All dataes show that a large diverse population of miRNAs may function to regulate gene expression in normal hepatocyte and hepatoma carcinoma cell.
