Purposes:In this experiment,papain was used to induce the formation of knee osteoarthritis in rabbits,and the effects of Xinzhiguzengshengwan on the levels of IL-10 (interleukin-10),TNF-α(tumor necrosis factor-α)MMP-3(matrix metalloproteinase-3),TIMP-1(tissue Inhibitor of Metalloproteinase-1)in serum of rabbits,the content of VEGF(vascular endothelial growth factor)in knee cartilage and the pathological changes of knee cartilage were observed,and the anti-inflammatory mechanism of Xinzhiguzengshengwan on knee osteoarthritis was discussed.To provide scientific and reliable experimental research basis for the treatment of knee osteoarthritis with traditional Chinese medicine.Material and Methods:Twenty-six healthy New Zealand rabbits were purchased and randomly divided into 19 rabbits in the modeling group and 7 rabbits in the blank group.Papain was injected into the knee cavity for modeling.After the modeling,one rabbit from each group was randomly selected for modeling identification.After successful modeling,the remaining 18 rabbits in the modeling group were randomly divided into model group,Xinzhiguzengshengwan treatment group and glucosamine positive control group,with 6 rabbits in each group and 6 rabbits in blank group.The Xinzhiguzengshengwan treatment group was given suspension 4ml/ Kg·d,the control group was given suspension of glucosamine hydrochloride 3.5ml/ Kg·d,and the blank group and model group were given normal saline intragastric administration 4ml/ Kg·d.Gavage was given once a day for 30 days.After the experiment,serum of rabbits were collected,and the contents of IL-10,TNF-α,MMP-3 and TIMP-1 in serum were detected by ELISA,VEGF content in knee cartilage was determined by immunohistochemistry,the changes of chondrocyte structure were observed by HE staining and Mankin’s score.Results:1.Elisa results:(1)Compared with blank group,serum IL-10 content in model group was decreased(P < 0.01);Compared with model group,serum IL-10 content in treatment group was increased(P < 0.01),serum IL-10 content in control group was increased(P < 0.05); Compared with the control group,the serum IL-10 content in the treatment group was increased(P < 0.01).(2)Compared with blank group,the contents of TNF-α and MMP-3 in serum in model group were increased(P < 0.01).;Compared with model group,the contents of TNF-α and MMP-3 in serum of treatment group and control group were down-regulated(P< 0.01).;Compared with the control group,the contents of TNF-α and MMP-3 in serum in the treatment group were down-regulated(P < 0.01).(3)Compared with blank group,the serum level of TIMP-1 in model group was increased(P < 0.01);Compared with model group,the serum level of TIMP-1 in treatment group and control group was increased(P < 0.01) Compared with the control group,the serum TIMP-1 content in the treatment group was increased(P < 0.01).2.Results of HE staining of articular cartilage: Compared with blank group,the score of model group was higher(P < 0.01).;Compared with model group,the scores of treatment group and control group were lower(P < 0.01);Compared with the control group,the score of treatment group was lower(P < 0.01).3.Immunohistochemical results:(1)The positive rate of VEGF cells was higher in the model group than in the blank group(P < 0.01);Compared with model group,the positive rate of treatment group and control group was lower(P < 0.01);Compared with the control group,the positive rate of treatment group was lower(P < 0.01).(2)Average absorbance of VEGF: compared with blank group,the average absorbance value of model group was higher(P < 0.01);Compared with model group,the average absorbance value of treatment group was lower(P < 0.01),while that of control group was lower(P < 0.05);Compared with the control group,the average absorbance value of the treatment group was lower(P < 0.01).Conclusion:1 Xinzhiguzengshengwan can promote the rehabilitation of knee osteoarthritis.2 The mechanism of action of Xinzhiguhyperplasia pills in the treatment of knee osteoarthritis is to reduce the content of IL-10 and TNF-α in serum,restore the balance ratio of MMP-3 and TIMP-1,and reduce the content of VEGF in cartilage,so as to reduce the inflammatory response,reduce the destruction of articular cartilage,and achieve the effect of the treatment of knee osteoarthritis.
