Purpose:To study the effect of Huatan Sanjie Chinese herbal compound on the expression of growth factor bFGF in nodular goiter rats,and to explore the mechanism of its treatment of goiter.And observe the treatment results of Chinese medicine compound Huatan Sanjie Chinese medicine compound,provide a theoretical basis for the treatment of goiter with Huatan Sanjie compound,and provide new ideas for clinical treatment of goiter.Material and method:A total of 120 SPF Wistar rats of 4 whole weeks of age and half male and half female were selected.After one week of feeding with conventional feed,30 rats were randomly selected as normal control group(NC)and fed with conventional feed throughout the course.Equal volume of deionized water was given daily.The other 90 rats were fed with low iodine diet,and were given sodium perchlorate for 19 days,followed by 2days of double distilled water,a total of 21 days for goiter model verification.After successful modeling,they were randomly divided into 3 groups,30 in each group,namely model control group(MC),levothyroxine sodium tablets group(L-T4),Huatan Sanjie compound group(HG).MC group was given equal volume of deionized water;h G was given an equal volume of Huatan Sanjie granule decoction converted by body surface area,that is,each rat was given 20.52g/kg of crude drug per day;L-T4 was given an equal volume of levothyroxine sodium tablet solution 0.2ug/kg;gavage once a day for a total of 77 days.The rats were sacrificed on the 21 st and 77 th days after the intervention factors were given,and 15 rats were sacrificed in each group.Abdominal aortic blood collection and thyroid tissue separation were performed by intraperitoneal anesthesia.The morphology of thyroid tissue in rats was observed by HE staining.TT3 and TT4 in serum were determined by RIA.TSH in serum was determined by IRMA.Thyroid index was calculated by dividing thyroid wet weight by body weight.The expression of bFGF was detected by immunofluorescence.Results:1.HE staining of thyroid cells: Compared with the normal control group,the thyroid cells of MC,HG and L-T4 changed,the follicular cavity of MC group disappeared,inflammatory infiltration existed,the morphology of epithelial cells changed,no double membrane structure was seen and the blood vessels around the follicles were abundant.Compared with the L-T4 group,the thyroid follicular epithelial cells in the HG group were flat and highly increased,which was significantly higher than that in the L-T4 group.2.Observation of thyroid function in each group: Compared with NC,TT3 and TT4 in other groups were significantly decreased and TSH was significantly increased on day 21.Compared with MC,the values of TT3 and TT4 in other groups were significantly increased,while TSH was significantly decreased.On the 77 th day,compared with NC,TT3 in HG group and L-T4 group increased significantly,while TSH in other groups still increased significantly.Compared with MC group,TSH was significantly lower and TT4 was significantly higher in HG group.3.Comparison of relative thyroid mass: On the 21 st day,the thyroid mass of the other groups was significantly higher than that of NC,P<0.05.On the 77 th day,compared with MC,the thyroid weight of the other treatment groups decreased significantly,P<0.05.HG group and L-T4 group were decreased,but the difference was not statistically significant,P>0.05.Conclusion:1.The Chinese medicine compound of resolving phlegm and resolving masses can treat nodular goiter by down-regulating the expression of growth factor bFGF in goiter rats.2.Compared with western medicine,Huatan Sanjie Chinese medicine compound can better down-regulate the expression of growth factor bFGF in the treatment of goiter,and more effectively inhibit the proliferation of thyroid cells under the premise of being friendly to the original thyroid cells. |
