Objective:To observe the effects of rolling-massager at different time points on local tissue morphology,tissue and serum levels of tumor necrosis factorα,interleukin 1βand nuclear factorκB signaling pathway-related indicators NF-κB p65 protein and inhibitor proteinαprotein of nuclear factorκB in rabbits with skeletal muscle injury,and to investigate the effect of rolling method on skeletal muscle injury time effect mechanism.Methods:Seventy-two New Zealand rabbits were selected and divided into a blank group,a model group,and a rolling method treatment group using a randomized grouping method,with24 rabbits in each group,and each group was further randomized into 6 subgroups(corresponding to different time points:24 h,3 d,5 d,7 d,9 d,and 11 d after injury),with 4rabbits in each subgroup.A model of acute blunt contusion was prepared by shaving the quadriceps muscle of the right lower limb of rabbits,drawing a 1cm~2circle on the quadriceps muscle belly about 3.5 cm above the patella,and striking the drawn circle 6 times with a homemade gravity hammer.group were bundled and treated at the same time point,and the treatment frequency was 1 time each at 10 am and 5 pm,3 min/time,for 3 days.Twenty-four hours after the end of the intervention,the morphological changes of the tissues were stained by hematoxylin-eosin(HE);the TNF-αand IL-1βlevels in the serum and damaged tissues were measured by enzyme linked immunosorbent assay(ELISA);Western blot(WB)was used to detect NF-κB p65 protein and IκBαprotein in damaged tissues.Results:HE staining results showed that compared with the blank group,inflammatory cell infiltration was obvious and edema increased in the model group,indicating successful modeling;inflammatory infiltration was further obvious in the 3d model group,and gradually stabilized at5-11 d.The inflammatory cell infiltration in the treatment group was intensified after 24 h of muscle injury by the rolling method,the muscle fiber fracture necrosis was more serious,the inflammatory response was strong,the cells were changed,the nuclei disappeared,and the inflammation in the 7 d rolling method treatment group was obviously improved with the best effect,and the difference with normal healthy muscle tissue was small.ELISA results:compared with the blank group,TNF-αin serum and TNF-αand IL-1βin tissues were significantly increased in the model group(P<0.01);TNF-αin serum and TNF-αand IL-1βin tissues were higher in the model group at 3 d than at 24 h.The 24 h rolling method treatment group expressed higher levels than the model group,and 7 d rolling treatment group was significantly lower than the model group(P<0.05);IL-1βexpression levels in serum were significantly higher in the model group compared with the blank group,and the levels in the rolling treatment group were lower than the model group(P<0.05).WB results showed that NF-κB p65 protein levels were significantly increased in the model group compared with the blank group(P<0.01);compared with the 24-h model group,the protein content of the 24 h rolling method treatment group was significantly reduced,but not statistically significant(P>0.05),and the remaining time points were reduced in all treatment groups,with the 7 d rolling method treatment group decreasing the most ratio(all P<0.05);while IκBαprotein was the opposite of NF-κB p65 IκBαprotein expression was completely opposite to NF-κB p65 protein expression,and IκBαprotein content in the model group decreased rapidly after modeling(P<0.01);compared with the 24 h model group,protein content in the rolling method treatment group increased,but it was not statistically significant(P>0.05);compared with the model group at the remaining time points,all treatment groups increased,and the ratio of increase was the most at 7 d(all P<0.05).Conclusions:1.24 h after skeletal muscle injury in rabbits was in the acute inflammatory phase,and the inflammatory factors did not decrease in the rolling method treatment group,indicating that the rolling method treatment was not effective at this time.2.The inflammatory situation began to gradually improve on the third day after muscle injury with rolling method treatment,and was most significant on the seventh day,indicating that this is the best treatment time point for rolling method intervention in skeletal muscle injury.Treatment at this time can fastest reduce the adverse effects of inflammatory factors,reduce apoptosis of skeletal muscle injury cells,accelerate the speed of skeletal muscle repair,and improve the quality of functional recovery.
