Observation On The Regulation Of Intestinal Flora And The Effect Of Simo Decoction On SCFAs In Rats With Heart Failur

Objective:To compare the difference of serum NT-proBNP and short-chain fatty acids(SCFAs),expression of hi stone deacetylase(HDAC)and inflammatory factor IL-6 in each group of rats and to explore the specific effects of SiMo Tang on intestinal microbiota structure and SCFAs in the treatment of heart failure rats.Methods:24 rats were randomly divided into model group,SiMo Tang group,eight captopril group and 10 blank group by intraperitoneally injection of 28 day isopropofol hydrochloride into heart failure rats.SiMo Tang group gave the SiMo Tang oral solution intervention,captopril group gave the captopril tablet intervention;The blank group and the model group were given saline.After 2 weeks of intervention,changes in intestinal flora and SCFAs in rat faeces were detected by 16Sr DNA sequencing and GC-MS.Serum levels of NT-proBNP,HDAC,and IL-6 were determined by ELISA in each group of rats.Results:Compared with the blank group,the model group rats prefer tranquility,are inactive,and respond slowly to external stimuli.In the later stage,Weight loss gradually occurs,food consumption is less,and the fur tends to lose luster,which even shows severe pleural effusion.In the model group,the left ventricular Ejection fraction(LVEF)decreased significantly(P<0.01),and the NT proBNP level increased significantly(P<0.01);The content of SCFAs,the metabolites of Gut microbiota,decreased(P<0.01),and the serum HDAC and IL-6 values increased(P<0.01).Compared with the model group,under the corresponding drug intervention,the overall condition of the rats in the SiMo Tang group and the Captopril group was lighter than the model group,the amount of activity of the rats was increased,the amount of food and water was more than the model group,the weight was slightly decreased,and the depilation symptoms were alleviated compared with the model group;The serum NT proBNP in SiMo Tang group and Captopril group decreased(P<0.01);The content of SCFAs increased(P<0.01);Both serum HDAC and IL-6 values decreased(P<0.01).Compared with the SiMo Tang group,the serum NT proBNP content in the Captopril group decreased(P<0.01),the SCFAs content decreased(P<0.01),the serum HDAC value increased(P<0.01),and the serum IL-6 value decreased(P>0.05).Overall,both the traditional Chinese medicine group and the Western medicine group can improve symptoms.Conclusion:SiMo Tang oral liquid can change the structure and quantity of Gut microbiota;It can promote the increase of SCFAs content in Gut microbiota,thereby inhibiting the production of HDAC and IL-6.
