Objective: Through experimental research,the expression characteristics of autophagy after spinal cord injury were clarified,and it was speculated that the effect of Jiaji Wenzhen on promoting the repair of spinal cord injury was achieved by affecting autophagy and inhibiting apoptosis.To explore the regulatory mechanism of Jiaji Wenzhen on autophagy-related factors in spinal cord tissue of rats with spinal cord injury.Methods: 72 rats were randomly divided into blank group,model group(spinal cord injury group),control group(3-Methyladenine 3-MA+Jiaji warm acupuncture treatment group)and observation group(Jiaji warm acupuncture treatment group).After successful modeling,each group of rats was given different interventions,and each group was divided into 7d and 14 d.The motor function of rats was evaluated by BBB score and inclined plate function score.The rats were sacrificed at 7d and 14 d.The spinal cord was taken for Nissl staining.The Bcl-2 protein was detected by immunohistochemical staining.The expression of LC3,Beclin1 and P62 protein was detected by Western blotting.Results: The BBB score and inclined plate function score of the normal group at each time point were higher than those of the other groups(P<0.05),and the observation group was higher than the model group and the control group(P<0.05).At each time point of Nissl staining,there were more residual neurons in the injured area of the treatment group,while the number of neurons in the model group and the control group decreased and the density of neurons was low.The results of Western blot showed that the expression of LC3 and Beclin1 protein in the observation group was higher than that in the model group and the control group,and the expression of P62 protein was lower than that in the model group and the control group.The results of immunohistochemistry showed that the expression of Bcl-2 protein in the observation group was higher than that in the model group and the control group at 7 d and 14 d after operation.Conclusion:1.Jiaji warm acupuncture can improve motor dysfunction in rats with spinal cord hemisection injury;the expression of Beclin1 and LC3 protein in spinal cord tissue of rats with spinal cord hemisection injury increased,the expression of P62 protein decreased,and autophagy was activated;2.3.Jiaji warm acupuncture can promote the occurrence of autophagy and inhibit apoptosis after spinal cord hemisection injury...
