Object:Apoptosis plays an important role in the pathogenesis of chronic obstructive pulmonary disease(COPD).This study attempts to determine whether Bufei decoction can improve the apoptosis level of COPD emphysema by regulating IL-17A/JNK/c-Jun pathway.Methods:A total of 42 clean grade ICR mice were randomly divided into blank group,model group,Bufei decoction group and methylprednisolone group,with 10 mice in each group.The mice model of COPD emphysema was simulated by self-made smoke tank for 90 days.After successful modeling,Bufei Tang Traditional Chinese medicine no-decoction granules and methylprednisolone tablets were injected into the stomach for intervention,and specimens were collected after continuous intervention for 15 days.The general state of the mice was observed during the experiment.The lung function of the mice was detected the next day after the last intragastasis,and pathological changes of the lung tissues were observed by HE staining;the expression of IL-17 A,JNK,p-JNK,and p-c-Jun in the lung tissues were detected by Western-Blot;immunohistochemical Cleaved Caspase-3 was detected.Lung tissue apoptosis was detected by TUNEL.Results:1.General condition: Mice in blank group are sensitive,quick in action,white and bright in hair.In the early stage of the model group,there was strong aggression,irritable temperament,slow action gradually appeared in the later stage of smoke exposure,partial hair removal,peeling,and thin body type.The symptoms of mice in Bufei decoction group and methylprednisolone group were improved compared with model group.2.HE staining: The size of the alveoli of mice in the blank group was normal and uniform,the structure of the alveoli was complete,and no emphysema was formed.In the model group,alveolar sizes varied,alveolar walls were mostly thin,fractured,or collapsed,part of alveolar walls were free,alveolar septa was ruptured,alveolar cavities were enlarged,and multiple alveoli were fused to form bullae.Compared with the model group,the number of enlarged alveolar walls in the Bufei decoction group and methylprednisolone group was lower,the alveolar walls were partially thinner and fractured,the alveolar cavities were enlarged,and bullae formation was occasionally observed in the bufei decoction group and methylprednisolone group.3.Lung function: Compared with blank group,FEV0.2/FVC,MMEF and PEF in model group,Bufei decoction group and methylprednone group were significantly decreased(P < 0.05).Compared with model group,FEV0.2/FVC and MMEF in Bufei decoction group and methylprednone group were improved(P < 0.05);There was no statistical difference in PEF.4.Lung tissue apoptosis was detected by TUNEL method: compared with blank group,the lung tissue apoptosis rate in model group was the highest,followed by Bufei decoction group and methylprednisolone group(P < 0.05).The apoptosis rates of Bufei decoction group and methylprednisolone group were lower than those of model group(P < 0.05).The apoptosis rate of methylprednisolone group was low in Bufei decoction group(P < 0.05).5.Immunohistochemical detection,Cleaved Caspase-3: Compared with blank group,Cleaved Caspase-3 protein expression levels in the model group,bufei-tang group,and methylprednone group were significantly increased(p < 0.05).Compared with model group,the histone expression levels of Bufei decoction group and methylprednisolone group were significantly decreased(p < 0.05).Compared with Bufei decoction group,the histone expression level of methylprednisolone was decreased(p <0.05).6.Western-Blot detection of the expressions of IL-17 A,JNK,P-JNK and p-c-Jun in lung tissue: compared with blank group,the expressions of IL-17 A,P-JNK and P-c-jun in model group and Bufei Tang group were significantly increased(P < 0.05);The expression of P-JNK in methylprednisolone group was higher(P < 0.05),but IL-17 A and p-C-Jun had no statistical difference.Compared with model group,IL-17 A,p-JNK and P-C-Jun in Bufei Tang group and methylprednisolone group were significantly decreased(P < 0.05).Compared with Bufei decoction group,the expression levels of IL-17 A,p-JNK and P-c-jun in methylprednisolone group were lower(P < 0.05).There were no significant differences in JNK expression among all groups(P > 0.05).Conclusion:1.Bufei Decoction can improve the alveolar structure,inhibit the apoptosis of alveolar epithelial cells and improve lung function in COPD emphysema mice.2.Bufei Decoction may reduce the expression of downstream apoptotic proteins by inhibiting the activation of IL-17A/JNK/p-c-Jun signaling pathway,thus alleviating lung tissue apoptosis.
