Objective: The effect of flavored Banxia Baishu Tianma soup on blood pressure was observed;To investigate whether the flavored Banxia Baishu Tianma decoction has anti-myocardial fibrosis effect,and whether it can improve myocardial fibrosis by inhibiting TGF-β expression,thereby inhibiting the HMGB1/TLR4 signaling pathway,and providing scientific evidence for revealing the mechanism of flavored Banxia Baishu Tianma decoction in the treatment of hypertension and hypertensive myocardial fibrosis.Methods:(1)20 16-week-old male SHR rats were divided into 4 groups according to the random number table method: low-dose group of Banxia Baishu Tianma Soup,high-dose group of Banxia Baishu Tianma Soup,model group,and losartan potassium group,with 5 rats in each group;Three more in the WKY group were set as a blank control group,for a total of 25 rats.(2)After 1 week of adaptive feeding,the blood pressure of SHR rats was detected by non-invasive smart sphygmomanometer at weeks 0,4 and 8;(3)At the end of feeding the 8th week,2 animals was randomly selected from each group and the myocardial tissue of SHR rats was observed by HE staining.The left ventricular mass fraction of SHR rats was calculated by measuring rat weight and heart weight.(4)Finally,3 in each group were selected for Western Blot experiments to detect the relative expression of TGF-β,HMGB1 and TLR4.Results:(1)Blood pressure assessment: Through the experiment on the effect of blood pressure on SHR rats,it was found that 4 weeks after administration,compared with the model group,the blood pressure level of the high-dose group and losartan potassium tablets group with flavored Banxia Baishu Tianma decoction was significantly reduced.Compared with losartan potassium group,the high-dose group with flavored Banxia Baishu Tianma decoction had a more significant antihypertensive effect.Eight weeks after administration,compared with the normal group,the blood pressure levels of the low,high-dose and losartan potassium groups of the flavored Banxia Baishu Tianma decoction were significantly reduced(P<0.05),and compared with the losartan potassium group,the blood pressure levels of rats in the high-dose group of the flavored Banxia Baishu Tianma decoction were significantly reduced(P<0.05).The results showed that both the flavored Banxia Baishu Tianma decoction group and the losartan potassium group could effectively reduce the blood pressure level of SHR rats,and the antihypertensive effect was the most obvious in the high-dose group of Banxia Baishu Tianma Soup,and the antihypertensive effect was better than that of losartan potassium group.(2)Left ventricular mass:After 8 weeks of drug intervention,the hearts of SHR rats were dissected,the rat heart weight and left ventricular weight were weighed and LVMI calculated,and the LVMI of rats in the flavored Banxia Baishu Tianma decoction group and losartan potassium group was significantly reduced(P<0.05),and there was no significant difference in the low-dose group of the flavored Banxia Baishu Tianma Soup,and compared with the losartan potassium group,the LVMI of SHR rats in the high-dose group of the flavored Banxia Baishu Tianma decoction was significantly lower than that in the model group(P<0.05).The hint showed that the heart weight index could be decreased in both the high-dose group of traditional Chinese medicine and the western medicine group,which could effectively reduce the LVMI of SHR rats and inhibit LVM,and the high-dose group of traditional Chinese medicine was better than that of western medicine.Myocardial tissue morphology:(3)The sections and HE staining of the myocardial tissue of SHR rats showed that the WKY control group had a smooth inner membrane,neatly arranged cardiomyocytes,normal structure,and no obvious collagen fiber hyperplasia.In the model group SHR rats,a large number of inflammatory cells were infiltrated,myocardial fibers were ruptured,and the myocardial cell space was significantly expanded,and a large number of collagen fibers could be seen.Different degrees of myocardial disorders,inflammatory cell infiltration,and different degrees of local collagen fiber arrangement disorders were seen in each intervention group.Among them,the high-dose group of traditional Chinese medicine had the most obvious improvement,suggesting that the flavored Banxia Baishu Tianma soup had a certain improvement effect on hypertensive myocardial fibrosis.(4)Protein expression: The expression of TGF-β,HGMB1,TLR4 and other proteins in SHR rats was detected by Western Blot method after a series of treatments on SHR rat myocardial tissue,and compared with the WKY control group,the expression of TGF-β,HMGB1 and TLR4 proteins in rat myocardial tissue in each model group was significantly increased(P<0.05).Conclusion: 1)Flavored Banxia Baishu Tianma soup has the effect of reducing blood pressure and anti-myocardial fibrosis in SHR rats.(2)TGF-β expression existed in SHR rats with myocardial fibrosis,and flavored Banxia Baishu Tianma decoction could reduce TGF-βexpression in SHR rats.(3)Flavored Banxia Baishu Tianma decoction can significantly downregulate the expression of proteins HMGB1 and TLR4 while inhibiting myocardial fibrosis;The mechanism may be related to the inhibition of HMGB1/TLR4 signaling pathway downtransmission.
