| Objective:To observe the effect of Danzikangxi Granule on the expression of autophagy-related protein in knee cartilage of KOA model rats by establishing knee osteoarthritis model and intervening with Danzikangxi Granule to explore the mechanism of Danzikangxi Granule in the treatment of KOA.Methods:45 male sprague-dawley rats were randomly divided into blank group(N=6)and model group(N=36)after adaptive feeding for 1 week.The model-making group was made according to the modified Hulth method,and then the model-making group was randomly divided into 6 groups,Danzikangxi Granule High,middle and low dose groups,celecoxib group,glucosamine sulfate group and model group and treated with corresponding drugs,the blank group and the model group were given 0.9%sodium chloride injection of the same volume by gavage.After 8 weeks of continuous intervention,X-ray examination was performed to observe the imaging features of the knee joint of the rats,and then the samples were taken to obtain the cartilage tissues of the knee joint for pathological staining,the expression levels of Becline1,LC3B and MMP-3 were detected by Western blot.Results:According to the X-ray film,the imaging manifestations of the knee joint of the rats in each dose group and the other two groups were improved to some extent compared with the model group(P<0.05).The cartilage tissues of the knee joint of rats were stained with HE and fuchsin-fixative green respectively to observe the pathological changes of cartilage.Compared with the blank group,the cartilage tissue of the model group was damaged seriously,the number of chondrocytes was decreased,the arrangement of chondrocytes was irregular and the level of chondrocytes was unknown.After the intervention of each drug group,the pathological condition of the cartilage of the knee joint of the rats in the model group was improved in different degrees,which showed that the number of chondrocytes was increased,the arrangement of chondrocytes was relatively orderly,the coloration was relatively uniform,and the cartilage layer was thicker than that in the model group.Compared with the model group,the expression levels of autophagy-related proteins Beclinl and LC3B in the articular cartilage of the rats in each dose group were significantly increased(P<0.05 or P<0.01),the expression of matrix metalloproteinase MMP-3 was significantly decreased(P<0.05).Conclusion:1.Danzikangxi Granule can reduce the degree of pathological injury and delay the degeneration of cartilage tissue in KOA model rats.2.Danzikangxi Granule could up-regulate the expression levels of autophagy-related proteins Beclinl and LC3B,and down-regulate the expression levels of MMP-3.3.Danzikangxi Granule may play a therapeutic role by increasing the level of autophagy and inhibiting the release of matrix metalloproteinase in the chondrocytes of KOA model rats. |
