Objective:Based on data mining and network pharmacology,to investigate the medication rules of external treatment of persistent infection in high-risk human papilloma virus(HR-HPV),and to analyze the molecular mechanism of action of drug treatment to guide clinical practice.Methods:1.Search for topical prescriptions of traditional Chinese medicine for the treatment of HR-HPV persistent infection based on CNKI,Wanfang,Weipu and other literature databases;The clinical records of patients with persistent HR-HPV infection who attended the Gynecology Clinic of Beijing University of Chinese Medicine from January 2018 to January 2023 were collected and sorted out,and prescriptions for external use of Chinese medicine were extracted.Excel was used to establish a literature database and a clinical database,and the prescriptions of traditional Chinese medicines that met the criteria were entered separately,and after unified and standardized processing,they were imported into the cloud platform of ancient and modern medical cases to obtain drug frequency,sexual taste,efficacy,and composition rules,etc.,and SPSS Modeler 18.0 was used to analyze the association rules between drugs and summarize the drug characteristics of external treatment of traditional Chinese medicine for the treatment of HR-HPV persistent infection.2.After the above data mining,high-frequency drugs were obtained for network pharmacology research.Using the TCMSP database of the TCMSP system pharmacology platform of traditional Chinese medicine,screen the active ingredients of compounds,Pubchem,Swiss target prediction and Uniprot protein databases were used to obtain drug targets,and OMIM database,TTD database,GeenCards database and DisGeNET database were searched to obtain disease targets.Cytoscape 3.7.1 was used to construct a visualization network,summarize the targets of drug intervention in diseases,construct protein interaction and bioconcentration analysis of key targets,explore the mechanism of action,and simulate and verify through molecular docking.Results:1.A total of 119 prescriptions were included in the literature database,and 605 prescriptions were included in the clinical database,and the following results were obtained(1).After data mining of the entered prescriptions:High-frequency drugs:in order of frequency of use,the top ten Chinese medicines in the literature are:phellodendron,matrine,borneol,comfrey,zedoary turmeric,cnidium monniere,rhizoma smilacis glabrae,rhizoma bletillae,alum,astragalus,and the top ten Chinese medicines used in the clinical part are phellodendron,baibu,comfrey,matrine,honeysuckle,old stork grass,cnidium monniere,peppermint,rhizoma smilacis glabrae,and honeycomb.(2).Nature,taste and belonging meridians:The medicinal properties of traditional Chinese medicines in the literature part and clinical part are mainly cold and warm drugs,the medicinal taste is mainly bitter,sweet and spicy,and the menstruation is attributed to liver meridian,spleen meridian and heart meridian,and the effect is mainly antipyretic and detoxifying drugs and clear heat and dry and wet drugs.(3).Association rules:The association rules in the literature section were the most associated with the pair of matrine and cnidium monniere,and the prescriptions using phellodendron and comfrey and matrine were tended to be compatible with honeysuckle;The analysis of the association rules of the clinical part showed that the association degree of phellodendron and comfrey was the highest,and the three flavor drugs of phellodendron,comfrey and old stork grass were closely related and were often used in combination.(4).After complex network analysis,some of the drugs prescribed externally in the literature were Phellodendron,Matrine,Comfrey,Zedoary turmeric,Borneol,Cnidium Monniere,and Outpatient Prescription Drugs were Phellodendron,Matrine,Comfrey,Old Stork Grass,Baibu and Honeysuckle.2.After network pharmacology research,phellodendron,matrine,comfrey,old stork grass,baibu and honeysuckle may act on p53,AKT1,VEGFA,TNF,IL6,MYC targets through active ingredients such as quercetin,luteol,kaempferol,β-sitosterol,etc.,GO enrichment analysis found that the drug is mainly involved in oxidation,apoptosis,phosphorylation and other biological processes,KEGG pathway enrichment analysis results show that PI3K-AKT,IL17 and other signaling pathways may be the main pathways for drugs to function.The molecular docking results showed that the core components and core targets had good binding ability.Conclusions:At present,traditional Chinese medicine external treatment for HR-HPV persistent infection often uses Chinese medicines such as phellodendron,matrine,comfrey,old stork grass,baibu and honeysuckle to clear heat and detoxify,dry the moisture from the body and stop the leucorrhea,which exert pharmacological effects through multiple components,multiple targets and multiple pathways to intervene in HR-HPV persistent infection,Intervention in HR-HPV persistent infection by improving body immunity,which can be used as a reference for further research in the later stage.
