Expression Of STAT3 Protein In Brain Tissue Of Rats With Acute Ischemic Stroke And The Regulatory Effect Of Early Acupunctur

ObjectiveTo evaluate the efficacy of scalp-body acupuncture therapy in the treatment of acute ischemic stroke by meta analysis.To observe the effects of scalp-body acupuncture on the expression of STAT3 protein and p-STAT3 protein in hippocampus of middle cerebral artery obstruction(MCAO)rats through animal experiments,and to explore the possible mechanism of acupuncture in treating acute ischemic stroke.MethodsStudy 1:The researchers retrieved randomized controlled clinical trials of acupuncture in the treatment of acute ischemic stroke from the database.The researchers screened literatures and extracted required data according to the exclusion criteria,evaluated the quality of the included literatures with reference to Cochrane bias risk assessment tool,and used RevMan 5.4 software for data analysis.Study 2:36 rats were divided into sham group(A,n=12),model group(B,n=12),and scalp-body acupuncture group(C,n=12).Every group was divided into 1d and 3d time window subgroups.MCAO model was prepared in group B and C,and sham operation model was prepared in group A.Group C was treated with scalp-body acupuncture,group A and group B were not treated,only fixed with fixator.After surgery,Zea Longa nerve function score was performed at 4h,Id and 3d.The expression of STAT3 protein and p-STAT3 protein were detected by immunofluorescence and immunohistochemistry.ResultsStudy 1:This study included 15 literatures and 1352 patients.Compared with the control group,scalp-body acupuncture combined therapy can improve clinical effective rate,reduce neurological deficits,improve motor function and daily living ability.Study 2:1.With the progression of disease,the neurological function score of group B was increased in 3d subgroup compared with 1d subgroup(P>0.05).After treatment,neurological function scores in group C showed a downward trend compared with group B(P>0.05).2.Immunofluorescence results showed that STAT3 protein was red and p-STAT3 protein was green,indicating co-localization of STAT3 protein and p-STAT3 protein.The fluorescence staining intensity of group A was weak.In group B,the fluorescence staining intensity increased significantly,p-STAT3 protein underwent nuclear transfer,and the green fluorescence was mainly concentrated in the nucleus.The fluorescence staining intensity of group C was lower than that of group B.3.Immunohistochemical results showed that the target protein was brown or yellow,and the expression of STAT3 protein was significantly higher in groups B and C than in group A(P<0.05).In group C,the expression level of protein in 3d subgroup was significantly lower than that in 1d subgroup(P<0.05).p-STAT3 protein expression:In 1d subgroup,the protein expression level of group B and group C was significantly higher than that of group A(P<0.05).In the 3d subgroup,the protein expression level of group B was significantly higher than that of group A(P<0.05),and the protein expression of group C was significantly lower than that of group B(P<0.05).In group B,the protein expression level of 3d subgroup was significantly higher than that of 1d subgroup(P<0.05).p-STAT3/STAT3:In 3d subgroup,the proportion of protein expression level in group B was significantly higher than that in group A(P<0.05),while the proportion of protein expression level in group C was significantly lower than that in group B(P<0.05).In group B,the proportion of protein expression level in 3d subgroup was significantly higher than that in 1d subgroup(P<0.05).Conciusion1.Meta analysis results showed that scalp-body acupuncture could reduce neurological deficits,improve motor function and daily living ability.2.In the early development stage of acute ischemic brain,phosphorylation of STAT3 protein to p-STAT3 protein is activated,which promotes nuclear translocation of p-STAT3 protein,and p-STAT3 protein enters the expression or transcription of nuclear regulatory genes.3.Scalp-body acupuncture can reduce the neurological function score and reduce the neurological function injury after cerebral ischemia in MCAO rats.4.Scalp-body acupuncture combined therapy can reduce the expression level of STAT3 protein and p-STAT3 protein,and inhibit the phosphorylation of STAT3 protein.This suggests that early acupuncture intervention can play a certain protective role in the brain,and the mechanism may be through inhibiting the STAT3-related pathway.
