Effect And Mechanism Of Shenqi Fuzheng Injection On Apoptosis And Autophagy Of Gastric Cancer Cell

Research Objective:In this experiment,the effects of Shenqi Fuzheng Injection on apoptosis and autophagy in vivo and vitro gastric cancer cells were analyzed from the molecular and cellular levels.The mechanism of apoptosis and autophagy was analyzed,so as to provide an experimental basis for the treatment of gastric cancer with Shenqi Fuzheng Injection,and also provide theoretical support for the treatment of gastric cancer with traditional Chinese medicine.Research Methods:1 The effect of Shenqi Fuzheng Injection on apoptosis and autophagy of gastric cancer cellsIn the first part of the experiment,human gastric cancer cell SGC-7901 was selected for culture to establish a gastric cancer cell model.The CCK-8 method was used to investigate the inhibitory effect of different concentrations of Shenqi Fuzheng Injection on the growth of gastric cancer cells SGC-7901.The concentrations of Shenqi Fuzheng Injection in each group were set to be 0,1,2,4,8,16 and 32g/L,and the intervention time was 48h.The optimal drug concentration of Shenqi Fuzheng Injection was 32g/L.The experiment was divided into two groups:control group(0g/L Shenqi Fuzheng Injection)and experimental group(32g/L Shenqi Fuzheng Injection intervention).The intervention lasted for 48 hours.The apoptosis level of each group was detected by Annexin V/PI staining.The expression of apoptosis-related proteins Bax and Bcl-2 was detected by Western Blot method.The Western Blot method was used to detect the differences in the expression of autophagy-related proteins Beclin-1 and LC3B in gastric cancer cells to observe the effect of Shenqi Fuzheng Injection on autophagy.2 The effect of Shenqi Fuzheng Injection on apoptosis and autophagy of gastric cancer transplant tumor cellsIn the second part of the experiment,female BALB/c nude mice were selected as research animals,and an animal model of subcutaneous transplantation tumor of gastric cancer cells SGC-7901 cells was established.The experimental animals were divided into two groups:the control group(0g/L Shenqi Fuzheng Injection)and the experimental group(32g/L Shenqi Fuzheng Injection).Firstly,by observing the volume and the mass of tumors,it was proved that Shenqi Fuzheng Injection has the effect of inhibiting the proliferation of gastric cancer cells.Furthermore,the expression of apoptosis-related proteins Bax and Bcl-2 was detected by Western Blot method,and it was found that Shenqi Fuzheng Injection could promote apoptosis.The expression of LC3B and Beclin-1 was detected by Western Blot method,and the effect of Shenqi Fuzheng Injection on autophagy of gastric cancer transplant tumor cells was explored.Research results:1 Shenqi Fuzheng Injection promotes apoptosis and autophagy of gastric cancer cells.CCK8 detection results:Shenqi Fuzheng Injections with concentrations of 1,2,4,8,16,and 32g/L have inhibitory effects on the growth of gastric cancer cells,and the difference was statistically significant(P<0.05),and it was concentration-dependent.The optimal drug action concentration of Shenqi Fuzheng Injection was 32g/L,and the optimal drug action time was 48h.Annexin V/PI staining to detect apoptosis:Compared with the control group,the apoptosis rate in the experimental group increased significantly,and the difference was statistically significant(P<0.05).Western Blot method to detect the apoptosis-related proteins expression:Compared with the control group,the expression level of Bax protein in the experimental group increased significantly,and the expression level of Bcl-2 protein in the experimental group significantly reduced,and the difference was statistically significant(P<0.05).Detection of autophagy-related protein expression by Western Blot method:Compared with the control group,the expression level of LC3B protein and the expression level of Beclin-1 protein in the experimental group also increased significantly,and the difference was statistically significant(P<0.05).2 Shenqi Fuzheng Injection promotes apoptosis and autophagy of gastric cancer transplant tumor cellsThe second part of the experiment successfully constructed a subcutaneous transplantation tumor model of gastric cancer cell SGC-7901 nude mice.Shenqi Fuzheng Injection has a significant inhibitory effect on the growth of gastric cancer transplanted tumors in nude mice.Compared with the control group,the volume and the mass of transplanted tumors in the experimental group were both significantly reduced,and the difference was statistically significant(P<0.05).Western Blot method to detect apoptosis-related protein expression:Compared with the control group,the expression level of Bax protein increased significantly and the expression level of Bcl-2 protein in the experimental group significantly reduced,and the difference was statistically significant(P<0.05).Detection of autophagy-related protein expression by Western Blot method:Compared with the control group,the expression level of LC3B protein and the expression level of Beclin-1 protein in the experimental group also increased significantly,and the difference was statistically significant(P<0.05).Conclusion and significance:1 Shenqi Fuzheng Injection has the effect of inhibiting the growth of gastric cancer cells in vitro and in vivo.2 Shenqi Fuzheng Injection inhibits the growth of gastric cancer cells by promoting apoptosis and autophagy,and its mechanism may be related to the expression of Bcl-2,Bax,Beclin-1,and LC3B.
