ObjectiveTo observe the effects of Jianpizhidong Decoction(JD)on connexin43(Cx43)and gap connexin30(Cx30)in cortex,striatum,hypothalamus and hippocampus of tic disorder(TD)mice model.MethodsMale ICR mice(n=60)were randomly divided into blank control(BC)group,model group,western medicine group and JD group(each n=15).TD model was established by intraperitoneal injectioning IDPN(350mg/kg/d)for consecutive 7 days.The rats were administrated by Tia(29.5 mg/kg)and JD(17.8 g/kg)respectively.The contents of Cx43 and Cx30 were assayed by immunohistochemistry,Western blot and RT-PCR separately.ResultsCompared with blank control group,the positive expression rates of Cx43 and Cx30 in cortex,striatum,hypothalamus and hippocampus of mice in model group were decreased to different degrees.Compared with model group,the positive expression rates of Cx43 and Cx30 in all drug groups were increased to varying degrees.Compared with blank control group,the expressions of Cx43 and Cx30 in cortex,striatum,hypothalamus and hippocampus of model group were decreased to different degrees.Compared with model group,the expression levels of Cx43 and Cx30 in cortex,striatum,hypothalamus and hippocampus of all drug groups were increased to varying degrees.The difference of Cx43 in the western drug group of cortex and model group was statistically significant(P<0.05),and the difference of Cx30 in the western drug group of cortex and hippocampus was statistically significant(P<0.05).The difference of Cx43 in cortex,striatum and hippocampus between TCM group and model group was statistically significant(P<0.05),and the difference of Cx30 in cortex and hippocampus between TCM group and model group was statistically significant(P<0.05).Compared with the western medicine group,the expression of Cx43 in the cortex,striatum and hippocampus of the Chinese medicine group showed an increasing trend,and the difference of Cx43 in the cortex and hippocampus of the Chinese medicine group and the Western medicine group was statistically significant(P<0.05).Compared with the western medicine group,the expression of Cx30 in the cortex and hippocampus of the Chinese medicine group showed an increasing trend,and the difference of Cx30 in the hippocampus between the Western medicine group and the Chinese medicine group was statistically significant(P<0.05).Compared with blank control group,mRNA expressions of Cx43 and Cx30 in cortex,striatum.hypothalamus and hippocampus of model group decreased to varying degrees.Compared with model group,the expression of Cx43 and Cx30 mRNA in cortex,striatum,hypothalamus and hippocampus of all drug groups increased to varying degrees,and the difference of Cx43 mRNA expression in cortex,striatum and hippocampus of Tipilidc group was statistically significant(P<0.05).The difference of Cx30 mRNA expression in the hippocampus and cortex of the western medicine group(i.e.,the Tipilide group)was statistically significant(P<0.05),and the difference of Cx43 mRNA expression in the cortex,striatum and hippocampus of the Jianpi Zhidang group was statistically significant(P<0.05).There were significant differences in Cx30 mRNA expression in cortex and hippocampus of Jianpi Zhidang group(P<0.05).Compared with Tipilide group.Cx43 mRNA expression levels in cortex,striatum and hippocampus Jianpi Zhidang decoction group showed an increasing trend,and the difference was statistically significant(P<0.05).Compared with Tiride group,mRNA Cx30 levels in the Jianpi Zhidang group in the cortex and hippocampus showed an increasing trend,and the difference was statistically significant(P<0.05).Conclusion The anti-tics mechanism of JD might attribute to increasing the expression of Cx43 and Cx30 in cortex,striatum,hypothalamus and hippocampus which could decrease the neurotoxicity of glutamate.
