| Purpose:Based on the theory of "Treating coronary heart disease from the spleen",this study aims to explore the effect mechanism of Jianpi Qvtan Huayu Prescription on AS rats with spleen deficiency and phlegm turbidity syndrome,and to provide theoretical and experimental basis for the treatment of AS.Material and method:Twenty SPF male SD rats were fed with adaptive feeding.After the rats were found to be normal after adaptive feeding,modeling was started.In addition to the normal group fed with normal diet,the other four groups were given high-fat diet to establish a rat model of spleen deficiency and sputum turbidity AS for 21 weeks.In parallel to feeding the two different diets,rats were randomized and given different intervention drugs for gavage therapy,A group daily give Jianpi Qvtan Huayu Prescription low dose lavage,B group was given the Jianpi Qvtan Huayu Prescription middle dose lavage,C group was given in Jianpi Qvtan Huayu Prescription by high dose lavage,the blank group and model group were given the same volume of normal saline by gavage,Vitamin D3 or saline were injected intraperitoneally on day 1,4,and 8 weeks,which took 21 weeks.During administration rats were monitored for weight and feces every two weeks.After administration,the pathological sections of the aorta of SD rats in each group were stained with oil red O to identify AS model.TC,TG,LDL,HDL,hs-CRP and ET were detected by enzyme-plate analyzer.Blood analyzer was used to detect the blood routine of rats.Open field test was used to test the average speed,total displacement,the number of penetration and the accumulated residence time of the central grid.Results:1.Oil red O staining results: Compared with the blank group,the staining in the model group was obvious and typical AS plaques were formed.After administration,the lipid staining area of aorta decreased to varying degrees,suggesting that the model of spleen deficiency and phlegm turbidity AS rats was successfully constructed,and Jianpi Qvtan Huayu prescription had significant improvement effect on AS.2.Body weight test results:Compared with blank group at the same period,the body weight of model group,group A,and group B were significantly increased(p<0.05),In the three-dose group of Jianpi Qutan Huayu Prescription,only the body weight of high-dose group C had no significant difference(p>0.05);Compared with model group,the body weight of rats in each group was significantly inhibited after medication(p<0.05),suggesting that jianpi Qutan Huayu Prescription could inhibit the weight growth of AS rats,and the inhibition effect of high-dose Jianpi Qutan Huayu Prescription was better.3.Fecal test results:Compared with blank group,total fecal weight and fecal water content in model group were significantly increased,and dry fecal weight was significantly decreased(p<0.05),but no significant differences were found in all fecal indexes in drug administration group(p>0.05).Compared with model group,the total fecal weight and fecal water content of rats in each group were decreased after administration,while the dry weight of feces was significantly increased(p<0.05),suggesting that the fecal volume and water content could be reduced by Jianpi Qvtan Huayu prescription,and the symptoms of loose stool could be improved.4.Open field test results: Compared with blank group,the average exercise speed of rats in model group was decreased,but there was no significant difference(p>0.05),and the average exercise speed of rats in each group was significantly increased after medication(p<0.05);Compared with model group,the average exercise speed of rats in each group was increased after medication,and there was a significant difference(p<0.05).Compared with blank group,the total exercise distance of model group was decreased,but no significant difference(p>0.05),and the total exercise distance of all groups was increased,but no significant difference(p>0.05).Compared with model group,the total movement distance of rats in group B and C was significantly increased after medication(p<0.05).There was no significant difference in the number of perforation among all groups(p>0.05).Compared with blank group,the total residence time of rats in central lattice was significantly increased in group A(p<0.05),while there was no significant difference in other groups(p>0.05).Compared with blank group,the activity area of model group was mainly corner and surrounding area,but the residence time of central area was longer,and the activity area of drug administration group was similar to blank group.Compared with model group,the activity of rats in each administration group mainly concentrated in the corner and surrounding area,and rarely passed through the central area.It is suggested that Jianpi Qvtan Huayu prescription can improve the activity degree of AS rats and improve the symptoms of fatigue and fatigue.5.Blood lipid test results: Compared with blank group,the contents of TG,TC,LDL and HDL in serum of model group and all drug administration groups were significantly increased(p<0.05);Compared with model group,the contents of TG,TC,LDL and HDL in serum of rats in all drug administration groups were significantly decreased(p<0.05),suggesting that Jianpi Qvtan Huayu prescription can improve the level of blood lipid in AS rats.6.Endothelial injury test results:Compared with blank group,the serum ET content in model group and all drug administration groups was significantly increased(p<0.05);Compared with model group,the content of ET in serum of rats in each group was decreased after treatment(p<0.05),suggesting that Jianpi Qvtan Huayu prescription can reduce the degree of endothelial injury in AS rats.7.Results of inflammatory factor detection: compared with blank group,the serum hs-CRP content in model group,group A,and group B were significantly increased(p<0.05),and there was no significant difference in serum hs-CRP content in high-dose C group among all groups(p>0.05);Compared with model group,h S-CRP content in each group was decreased after medication(p<0.05),suggesting that Jianpi Qutan Huayu prescription could reduce the inflammation level of AS rats,and the anti-inflammatory effect of high-dose Jianpi Qutan Huayu prescription was better.8.Safety evaluation: There were no adverse effects in all group during the experiment,and there were no significant differences in the number and changes of blood routine indexes.Suggests that Jianpi Qvtan Huayu prescription It is safe.Conclusion:Jianpi Qvtan Huayu Prescription can intervene in the rats AS the formation of plaque,reduce weight of rats,improve indicators and behavior reaction of excrement and urine,improve lipid levels,reduce the hs-CRP and the synthesis and release of ET,reduce vascular inflammation and protect the vascular endothelial cell structure,and Jianpi Qvtan Huayu Prescription has good safety,can control and treat the occurrence and development of AS.and provides experimental basis for "treating coronary heart disease from spleen"... |
