Purpose:A model of RSV pneumonia in young rats infected with respiratory syncytial virus(RSV)infection was established,and the expression of TLR4,NF-κB p65,AQP1,AQP5 proteins and related factors in the RSV-induced RSV pneumonia model was explored by lung clearing pectoral cream.Material and method:RSV virus(respiratory syncytial virus long strain)was used for 3 consecutive days to establish and evaluate the RSV pneumonia SD juvenile rat model,observe the general state of rats,lung tissue pathology,calculate lung index,and after the model was successfully established,the next step of the experiment was carried out.63 SD juvenile rats were randomly divided into 3 groups,normal group,model group,lung clearing chest cream group,except for the normal group,the other two groups were nasal RSV virus,and the RSV pneumonia model was established.Observe the general state of the rats.On the 3rd,5th and 7th days of administration,each group randomly executed 7 rats for sampling,measured body weight and lung weight,calculated the lung index of each group,observed the lung pathological state of rats by HE staining,and detected the expression of TLR4,NF-κB p65,AQP1 and AQP5 proteins in rat lung tissue by Westernblotting,and the levels of IL-6,IL-1β,TNF-α,IFN-γ in serum were detected by ELISA method.Results:1.After rhinoplasty in RSV pneumonia rats,the general state is not good,mental atrophy,diet water reduction,weight loss,reduced mobility,hair dullness;after dissection,the lung tissue of the model group is visible to the naked eye,there are pathological manifestations such as edema and congestion;the lung index becomes larger and differs from the normal group(P<0.05);HE staining can show interstitial edema,lung tissue structure disorder,epithelial cell damage,a large number of inflammatory cells infiltrated,and the alveoli are destroyed,compared with the normal group,the pathological score of the model group is higher than that of the normal group(P<0.05),suggesting that the RSV pneumonia model model is successfully molded.2.HE staining of lung tissue found that after RSV infection,rat lung tissue,the model group appeared obvious lung tissue structure disorders,exudation of inflammatory cells,alveoli were destroyed,alveolar hyperplasia,interstitial edema.Cell hyperplasia,infiltration of inflammatory cells,edema of the interstitium.The inflammatory cells in the lung clearing pectoral cream group were significantly improved,and only a small number of inflammatory cells were destroyed,and the destruction of the alveoli was not obvious.After infection,the lung index and pathological score,and the lung clearing group were different from the normal group and the model group(P<0.05).3.The results of TLR4,NF-κ B p65,AQP1 and AQP5 by Western blot method showed that the lung clearing group had differences compared with the model group(P<0.05).Compared with the normal group,the expression of TLR4 and NF-κ B after RSV infection on days 3,5 and 7 of the model group was increased(P<0.05),and peaked on day 7,while the expression of AQP1 and AQP5 was decreased(P<0.05),and the expression was minimized on day 7.Compared with the normal group,the expression of TLR4 and NF-κB in the lung clearing pleoplasm group was higher than that in the normal group,and the expression of AQP1 and AQP5 was lower than that in the normal group,and the expression of TLR4 and NF-κB was downregulated(P<0.05)compared with the model group,and the expression of AQP1 and AQP5 increased.4.El ISA method was used to detect the serum levels of IL-6,IL-1β,TNF-α,IFN-γin the time points of each mouse in 3 groups of rats,and the results showed that compared with the normal group,the serum IL-6,IL-1β,and serum of the mice in the model group at each time point were compared with the normal group.TNF-αexpression was significantly increased(p<0.05),peaked on the seventh day,IFN-γexpression was significantly reduced(p<0.05),INF-γ was lowest at day 7;compared with the model group,the serum expression of IL-6,IL-1β,TNF-α at all time points was reduced,and the expression of IFN-γ was upregulated;compared with the normal group,the serum expression of IL-6,IL-1 β,TNF-α in the lung-clearing thoraphatic group was increased at all time points.IFN-γ express downcondition.Conclusion:1.Lung clearing pectoral cream group can effectively alleviate the abnormal manifestations of RSV pneumonia model and alleviate the inflammatory lesions of its lungs.2.The TLR4/NF-κB signaling pathway is involved in the inflammatory response of RSV pneumonia in young rats.3.Lung clearing thoracoplasm may exert its anti-RSV effect by inhibiting the TLR4 /NF-κB signaling pathway,inhibiting the production of inflammatory cytokines.
