Objective: To investigate the possible mechanism of fucong tablet delaying hearing loss in aged C57BL/6J mice by observing the changes of calcium binding protein content in cochlear nucleus(CN),and to provide theoretical basis for the prevention and treatment of senile deafness by Fucong tablet.Methods:Twenty-four 6-month-old C57BL/6J mice were randomly divided into two groups after ABR detection,the Fucong tablet treatment group and the elderly blank control group,which were fed with the diet containing Fucong tablet and ordinary diet for 6 months,respectively.Twelve 1-month-old C57BL/6J mice were selected as the young blank control group at the end of feeding.Auditory brainstem response audiometry(ABR)was used to evaluate the hearing status of each group.The expression of Nite and calcium binding protein(PV,CR,CB)in CN neurons of the three groups were observed by Nissl staining and IHC.Results:1.The average hearing threshold of the young blank group(1 month old)was significantly lower than that of the old blank group(6 months old)and(12 months old),and there were significant differences between the thresholds of 1 month old and 6 and12 months old(P<0.01).The 12 month old blank group and fucong tablet group showed different degree of hearing threshold elevation,the hearing threshold of mice in the aged blank group increased more obviously and the comparison of hearing threshold between the two groups was statistically significant(P<0.05).2.Comparison between the young blank group and the old blank group: The contents of nissl bodies,CB and CR in CN neurons of the old group were significantly lower than those of the young blank group,the content of PV was higher than that of the young blank group,the differences were all statistically significant(P<0.05),and the contents of nissl bodies and CB were significantly different(P< 0.01).3.Comparison between Fucong tablet treatment group and elderly blank group:the content of nissl bodies,CB and CR in CN neuronsof the Fucong group was higher than that of the elderly blank group,PV was lower than that of the elderly blank group.Only the content of nissl bodies and CB was significantly different between the two groups(P<0.05),CR and PV had no significant difference((P>0.05).Conclusion:Hearing loss in aged C57BL/6J mice was accompanied by damage of cochlear nucleus neurons,decrease of CB and CR,and increase of PV.Fucong tablets can effectively delay the hearing loss,and the mechanism may be related to the inhibition of calcium binding protein D-28K(CB)reduction and the delay of cochlear nucleus neuronal decline. |
