Objective:To establish a mouse model of dry eye with liver-kidney yin deficiency,and to explore the therapeutic effect and mechanism of acupoint catgut embedding on mice with dry eye and liver-kidney yin deficiency.Methods:60 8-week-old C57 female mice were selected,10 mice were randomly selected as the normal group,and the remaining 50 mice were placed in an electronic moisture-proof box for 4 weeks,and long-term irritated for 3 weeks at the same time to establish a dry eye liverkidney-yin deficiency mouse model.After successful modeling,they were randomly divided into model group,testosterone group,catgut embedding group,sham embedding group,and acupuncture group,Acupuncture treatment,the cycle is 4 weeks.During the period,the general state,tear secretion and corneal fluorescence staining of the mice in each group were dynamically observed,once every 2 weeks.Mice were sacrificed immediately after 4 weeks of intervention,and the lacrimal glands and cornea were removed,and paraffin sections were made to observe their pathological morphology by HE staining,and immunohistochemical staining to observe the inflammatory reaction.Orbital blood was collected to collect blood,and the levels of testosterone and estradiol in serum were measured.Results:1.Observation of rectal temperature,body weight and general state of miceAfter 4 weeks of treatment,the rectal temperature of the mice in the catgut embedding group and the acupuncture group was significantly lower than that in the model group(P<0.05),and the difference was statistically significant.After 4 weeks of treatment,compared with the model group,the weight of mice in the catgut embedding group,the acupuncture group and the testosterone group increased significantly compared with the model group,and the difference was statistically significant(P<0.01).After treatment,the general state of the mice in the catgut embedding group and the acupuncture group improved slightly,with thicker hair,more flexible responses,increased activity,and reduced screaming and biting frequency.2.Mouse tear secretionAfter 2 weeks of treatment,the amount of tear secretion of the mice in the catgut embedding group was higher than that before treatment.After 4 weeks of treatment,there was no significant difference in tear secretion between the catgut embedding group and the normal group,but there was a significant difference compared with the model group(P<0.01).3.Mouse tear film break-up timeAfter 2 weeks of treatment,the tear film break-up time of the mice in the acupuncture group was slightly increased compared with that before treatment;after 4 weeks of treatment,compared with the mice in the model group,the tear film of the mice in the catgut embedding group,the acupuncture group and the testosterone group was significantly higher.The rupture time was significantly increased(P<0.01).4.Image Analysis of Mouse Cornea Fluorescent StainingAfter 2 weeks of treatment,compared with the model group,the corneal fluorescence staining scores of the catgut embedding group and the testosterone group were significantly lower(P<0.01).After 4 weeks of treatment,the corneal staining of the mice in the catgut group,the acupuncture group and the testosterone group was relieved,and the corneal damage was improved;The corneal fluorescence staining scores of mice were significantly decreased(P<0.01).5.HE staining of mouse cornea and lacrimal gland tissueCompared with the mice in the model group,the corneal tissue structure integrity and cell morphology of the mice in the catgut embedding group,the acupuncture group and the testosterone group were significantly improved,the cells were arranged more regularly,the corresponding damage was alleviated,and the cells had different degrees of edema.Compared with the mice in the model group,the lacrimal gland tissue acinar structure and morphology of the mice in the catgut embedding group,the acupuncture group and the testosterone group were significantly improved,and the size of the acinus was more uniform.6.Immunohistochemical staining of mouse lacrimal gland tissueCompared with the mice in the model group,the expression of inflammatory factors in the catgut embedding group,the acupuncture group and the testosterone group decreased,and the color area decreased.Compared with the model group,the expressions of IL-1,IL-6and IL-8 in the catgut embedding group were significantly different(P<0.05),and the expressions of IL-6 and IL-8 in the acupuncture group were also significantly different(P<0.05).7.Mouse sex hormone detectionThere was no significant difference in the levels of estradiol among the groups;compared with the normal group and the model group,the serum testosterone content in the testosterone group was significantly increased(P<0.01).The testosterone content of catgut embedding group was closer to the testosterone content of testosterone group than other groups.Conclusion:Acupoint catgut embedding can improve rectal temperature,body temperature and symptoms and signs in mice with dry eye and liver-kidney yin deficiency,increase tear film break-up time,improve corneal injury,and restore corneal and lacrimal gland tissue morphology.The release of IL-6 and IL-8 reduces the inflammatory response and is related to the regulation of testosterone levels.
