Objective: The purpose of this experiment is to investigate the effects of Fangtuo Yufa essence on the regulation of testosterone,dihydrotestosterone,hepatocyte growth factor level,and hair follicle microenvironment in local skin tissue of SD androgen-derived alopecia rats to clarify the relevant mechanism of action of Fangtuo Yufa essence,and to provide an objective basis for the promotion of its use in clinical practice.Methods: For six weeks,the rat model of androgenic depilation was established by subcutaneous testosterone propionate injection(5mg/kg /d).Meanwhile,the drug was evenly applied to the selected observation area of the hair removal experiment.During the experiment,the general state of rats was observed regularly.The local skin microenvironment,the number and shape of hair follicles,the histopathological changes of hair follicles and sebaceous glands,the expression of hepatocyte growth factor,testosterone,and dihydrotestosterone were detected utilizing dermoscopy,pathological sections,immunohistochemistry,Western blot,and ELISA.Results:1.Observation results: During the experiment,the increasing trend of body weight and hair quality among the groups were the same,with no statistical significance(P>0.05).Generally,it can be seen that the groups injected subcutaneously with testosterone propionate injection have different degrees of swelling,bulge,scratch,blood scab,and erosion.In the blank group and control group,the hair was thicker in the skin group.The hair was sparse and greasy in the model group,and the hair follicle mouth dilated;compared with the Minoxidil Tincture group and the Fangtuo Yufa essence group,the yellow spots were obvious.The "Mao Zhou" sign was seen in the Minoxidil Tincture group and the Fangtuo Yufa essence group for 4-5weeks.2.test results: The Fangtuo Yufa essence group can significantly reduce testosterone and dihydrotestosterone levels in the skin tissue of androgenic depilation rats;the content of testosterone was159.84 ±10.685pg/ml,dihydrotestosterone content was 5.635 ±0.322nmol/L,the difference was significant compared with the model group(P<0.01).The Fangtuo Yufa essence group was better than the minoxidil group in reducing the level of dihydrotestosterone in the local skin(P<0.05).The Fangtuo Yufa essence group could increase hepatocyte growth factor in local skin tissues(P<0.05).By HE staining under a light microscope(100 times),compared with the model group,Minoxidil Tincture group and the Fangtuo Yufa essence group could increase the number of local skin follicle density and improve the miniaturization of the hair follicle,the number density of hair follicles in Minoxidil Tincture group was 31.33 ±7.280,is greater than the Fangtuo Yufa essence group(P<0.05).Conclusion: The Fangtuo Yufa essence can effectively reduce testosterone and dihydrotestosterone content in local skin tissue of androgenic depilation rats,promote the expression of hepatocyte growth factor in local skin tissue,improve the local hair follicle microenvironment,increase the number of fo llicle density,and improve the miniaturization of hair follicles.
