Objective:To investigate the effects of Yiqi Huoxue prescription on Nrf2(recombinant and synthetic protein)and NQO1(quinone oxidoredutase)expression and oxidative reaction in rats with cerebral ischemia-reperfusion injury.Methods:Forty SPF male SD rats were selected and randomly divided into sham operation group(n = 10)and model preparation group(n = 30)by random number table method after adaptive feeding for one week.Model preparation group established the rat middle cerebral artery ischemia/reperfusion model(MCAO/R)by wire embolization method,and then selected successful rats were divided into model group,Yiqi huoxue prescription group and Western medicine group by random number table method,each group contained 10 rats.Sham operation group and model group were given equal volume of distilled water and equal volume of normal saline by intraperitoneal injection;Chinese medicine group was given equal volume of Yiqi huoxue prescription and equal volume of normal saline by intraperitoneal injection;Western medicine group was given equal volume of distilled water and equal volume of edaravone injection by intraperitoneal injection.M-nss score was used to evaluate the degree of neurological impairment in rats.Hematoxylin eosin(HE)staining was used to observe the morphological changes of brain cells.Western Blot was used to detect Nrf2 and NQO1 expression in cerebral ischemia tissue.SOD activity,GSH content,MDA content and NO content in cerebral ischemic tissue were determined by the kit.The experimental data were expressed as mean ± standard deviation(X ± S).LSD method was used for pairwise comparison with homogeneity of variance,and Games-Howell method was used for non-homogeneity of variance.P < 0.05 indicated that the difference was statistically significant.Results:1.Neurological function score: there was no neurological deficit in the sham operation group.Compared with the model group,the neurological deficits of MCAO/R rats were significantly improved in both the TCM group and the WESTERN medicine group(P < 0.05),and there was no significant difference between the TCM group and the Western medicine group(P > 0.05).2.HE staining: The cortical neurons in the sham operation group were not damaged,and the nerve cells were clear in outline,orderly in arrangement,intact in structure,and uniformly stained.The model group was characterized by uneven staining in ischemic area,degeneration and shrinkage of nerve cells,nuclear pyknosis,destruction of cell structure,chromatin edge collection,and formation of a large number of vacuoles.Compared with the model group,the pathological changes of nerve cells in the traditional Chinese medicine group and the western medicine group were relatively lighter,the number of cells with good shape increased,the vacuole decreased,and the cell contour was clearer.3.Expressions of Nrf2 and NQO1: Compared with the sham operation group,the expressions of Nrf2 and NQO1 in the model group,the TRADITIONAL Chinese medicine group and the Western medicine group were increased at all time phases(P < 0.05).Compared with model group,Nrf2 and NQO1 expressions in TCM group and WESTERN medicine group were significantly increased(P < 0.05).There was no difference in Nrf2 and NQO1 expression between Chinese medicine group and Western medicine group(P > 0.05).4.Oxidative stress related indexes: Compared with sham operation group,MDA content and NO content in model group were significantly increased,while SOD content and GSH content were significantly decreased(P < 0.05).Compared with model group,MDA content and NO content in TCM group and WESTERN medicine group were significantly decreased,while SOD content and GSH content were significantly increased(P < 0.05).There was no difference in oxidative stress index between Chinese medicine group and Western medicine group(P > 0.05).Conclusion:1.After is CHEMIa-reperfusion in rats,yiqi huoxue can significantly improve the symptoms of neurological deficit in rats,reduce the score of neurological deficit,increase the expression of Nrf2 and NQO1 proteins in the brain tissue of rats,inhibit the oxidative stress response,and thus alleviate the cerebral ischemia-reperfusion injury in rats.2.Both Yiqi huoxue Prescription and edaravone can alleviate cerebral ischemia-reperfusion injury in rats,and there is no significant difference in their therapeutic effects. |
