Effect Of Annaopingchong Prescription On Bax/Bcl-2 Protein Expression And Neuronal Apoptosis After Cerebral Hemorrhag

Objective: To study the effect of Annaopingchong Decoction on the expression of Bax and Bcl-2 protein in rats after intracerebral hemorrhage,and to explore the mechanism of its action on nerve cell apoptosis after intracerebral hemorrhage.Methods: Healthy SD rats were randomly divided into sham operation group(Group J)and intracerebral hemorrhage model group.After successful modeling,intracerebral hemorrhage rats were randomly divided into model group(group M),annaopingchong decoction group(group A)and oxiracetam group(group O).The four groups were set at three time points of 1d,3d and 7d to ensure 8 rats in each group.According to the conversion method of body surface area equivalent dose between human and animal,the group of annaopingchong decoction and the group of oxiracetam were given corresponding drugs and corresponding doses by gavage;The model group and sham operation group were perfused with distilled water.Each group completed the neurological function score according to the corresponding time points.After the brain tissue was taken,TUNEL staining was performed,and the expression of Bax and bcl-2 protein was measured by Western blot.Results: 1.Neurological function score: compared with the sham operation group,the scores of model group and drug group at each time point were significantly higher,and the difference was statistically significant(P < 0.05);Compared with the model group,the score of the drug group decreased significantly at each time point,and the difference was statistically significant(P < 0.05);The score of annaopingchong decoction group 7 days after intracerebral hemorrhage was lower than that of oxiracetam group(P < 0.05).2.TUNEL staining: compared with the sham operation group,the apoptosis rate of model rats at different time points was significantly higher than that of the sham operation group(P < 0.05).3.Western blot test:(1)Bax protein expression: Bax protein in sham operation group was significantly lower than that in intracerebral hemorrhage model group on 1d,3d and7d(P < 0.05);The expression of Bax protein in annaopingchong decoction group and oxiracetam group was significantly lower than that in model group at each time point(P < 0.05);There was no significant difference between drug groups at different time points(P > 0.05).(2)Bcl-2 protein expression: Bcl-2 protein in sham operation group was significantly higher than that in intracerebral hemorrhage model group on 1d,3d and7d(P < 0.05);Compared with the model group,the expression of Bcl-2 protein in annaopingchong decoction group and oxiracetam group was up-regulated at each time point,and the difference was statistically significant(P < 0.05);There was no significant difference between drug groups at different time points(P > 0.05).(3)Bax / Bcl-2 ratio: compared with the sham operation group,the Bax / Bcl-2value of 1d,3d and 7d groups was significantly up-regulated,and the difference was statistically significant(P < 0.05),while the Bax / Bcl-2 value of annaopingchong decoction group and oxiracetam group was significantly down regulated compared with the model group(P < 0.05).Conclusion: 1.Annaopingchong recipe can reduce the symptoms of neurological deficit after intracerebral hemorrhage in rats,inhibit cell apoptosis and play a protective role in brain tissue.2.The ratio of Bax / Bcl-2 in intracerebral hemorrhage model group increased.Annaopingchong Decoction group can down regulate the expression of Bax protein,up regulate the expression of Bcl-2 protein,reduce the ratio of Bax / Bcl-2,inhibit cell apoptosis after intracerebral hemorrhage to a certain extent and reduce secondary brain injury.
