| Objective: By comparing the changes of IL-33 and IL-36 levels in the serum of psoriasis-like mouse models after treatment with different doses of Yinxieping Pills,the therapeutic effect of Yinxieping Pills was studied.Methods: According to the random number table,48 clean and sterile BALB/c mice were divided into 6 groups,the blank control group,the model control group,the Acitretin capsule group and the different dose groups of Yinxieping pills,namely high,medium and low dose groups,8 per group.The back hair of 48 experimental mice was shaved with a shaver,with an area of about 3cm×4cm.Except for the blank control group,the shaved area on the back of the other groups of experimental mice were all treated with 5% imiquimod cream.Evenly externally coated,mice in the blank control group were modeled with petroleum jelly,once a day,for 8consecutive days.At the same time,the mice in each group were given intragastric intervention.The low,medium and high dose groups of Yinxieping Pills were given intragastric administration with 0.4ml of Yinxieping Pill powder diluted solution of different concentrations.The number of intragastric administration was once a day,and the intragastric administration was continuous.8 days.Gavage the blank control group and the model control group with 0.4ml of normal saline;gavage the acitretin capsule group with0.4ml of acitretin diluent solution;observe the experiment with naked eyes and record with a professional camera during the experiment Skin lesions on the back of mice.Twenty-four hours after the last medication,the skin lesions were scored according to the PASI scoring standard.About 1.0 m L of blood was collected from the eyeball,and then the levels of IL-33 and IL-36 in the serum obtained in this experiment were detected by ELISA.The skin lesions of diseased mice were taken,sliced,and HE stained to observe the pathological changes.Results:(1)Compared with the blank control group,the PASI scores of the mice in the low,medium and high dose groups of Yinxieping Pills,the Acitretin capsule group and the model control group were significantly increased(P<0.05);compared to the model In the control group,the PASI scores of the tested skin lesions of the mice in the low,medium and high dose groups of Yinxieping Pills,and the Acitretin capsule group were significantly decreased(P<0.05).(2)Compared with the blank control group,the IL-33 and IL-36 levels in the serum of the model control group increased significantly(P<0.05);compared to the model control group,the IL-33 and IL-36 levels in the serum of the Acitretin capsule group Significantly reduced(P<0.05);Compared with the model control group,the levels of IL-33 and IL-36 in the serum of the three different dose groups of Yinxieping Pill,low,medium and high,were significantly reduced(P<0.05).(3)Compared with the model control group,the pathological changes of the skin lesions in the low,medium and high dose groups of Yinxieping Pill were reduced to varying degrees.conclusion:1.The PASI score of the experimental mice in the Yinxieping Pill group decreased,indicating that Yinxieping Pill has a good therapeutic effect on psoriasis.2.The serum levels of IL-33 and IL-36 in the model control group were significantly higher than those in the blank control group,suggesting that IL-33 and IL-36 are related to the pathogenesis of psoriasis.The m RNA level and protein level of Notch1 and caspase3 of the model control group were significantly increased suggested that may be involved in the pathogenesis of psoriasis3.The expression levels of IL-33 and IL-36 in the serum of the low,medium and high dose groups of Yinxieping Pills were significantly decreased,suggesting that the mechanism of Yinxieping Ping Pills in the treatment of psoriasis may be through inhibiting the expression of IL-33 and IL-36 achieve. |
