Objective: To explore the expression level and clinical significance of peripheral blood micro RNA-150-5p(miR-150-5p)in patients with chronic obstructive pulmonary disease(COPD)complicated with pulmonary hypertension(PH).Methods: A total of 60 patients with stable COPD in the department of the Third Hospital of Changsha from July 2019 to December 2020 were selected and divided into two groups according to systolic pulmonary arterial pressure(SPAP): COPD with PH(30 cases)and COPD without PH(30 cases).20 healthy persons were selected as control group at the same period.The general data of all subjects,including sex,age,Body Mass Index(BMI),N-Terminal Pro-brain natriuretic peptide(NT-proBN)were collected and the expression level of miR-150-5p in peripheral blood was detected.The general data and the difference of miR-150-5p expression level in the three groups were compared,the correlation between miR-150-5p expression level and the level of NT-proBNP and SPAP was analyzed,the diagnostic efficacy of peripheral blood miR-150-5p in COPD patients complicated with PH was evaluated.Results:.(1)There was no significant difference in sex,age and BMI among the three groups(P>0.05).The level of NT-proBNP in COPD patients with PH was the highest(533.95±139.03 pg/ml),followed by COPD patients without PH(148.91±49.47 pg/ml),and lowest in healthy people(35.29±11.40 pg/ml).There were statistical differences among the three groups(P<0.01).(2)The expression of miR-150-5p in COPD with PH group(0.144±0.069)was lower than that in COPD without PH group(0.345±0.096)and the healthy controls(0.556±0.241)(P<0.01).At the same time,The expression of miR-150-5p in COPD group was lower than that in control group(P<0.01).(3)Analysis of the receiver operating characteristic(ROC curve)showed that the area under ROC curve was 0.982±0.012,and 95%confidence interval(CI)was 0.958~1.000.It showed that miR-150-5p could predict the PH of COPD with high accuracy.(4)The correlation analysis indicated that the relative expression of miR-150-5p was negatively correlated with NT-proBNP and SPAP,respectively(r=-0.872,P<0.01;r=-0.884,P<0.01).Conclusion: The expression of peripheral blood miR-150-5p in COPD patients with PH decreased,which was negatively correlated with NT-proBNP and SPAP.It may be considered as a potential biomarker for early diagnosis and evaluation of secondary PH in COPD patients.
