| Objective:To investigate the effect of berberine on nasopharyngeal carcinom CNE2 cell migration and its related mechanism.Methods:1.Nasopharyngeal carcinoma CNE2 cells were treated respectively with berberine at different concentrations(0,2.5,5,10,20,40μmol/L)and fluorouracil at different concentrations(1,2,4,8,16μg/ml)for 24、36、48hours,and the cell proliferation was detected by MTT assay.The appropriate experimental concentration was determined.2.The effects of 5、20μmol/L berberine group、fluorouracil positive control group、blank control group and 12.5μmol/L Sc79group、20μmol/L berberine group、Sc79 combined with berberine group,25μmol/L LY294002 group、blank control group on the migration of nasopharyngeal carcinoma CNE2 cells were detected by scratch test and transwell test.3.The protein expression levels of PI3K,Akt,p-Akt,N-cadherin,E-cadherin and β-catenin were detected by WesternBlot.Results:1.Berberine could inhibit the proliferation of nasopharyngeal carcinoma CNE2 cells at 0,2.5,10,20,40μmol/L in a dose-and time-dependent manner.There were significant differences among 24h,36h and 48h groups(P<0.01).2.After intervention for 36hours,the scratch test of each group showed that the mobility of 20 μmol/L berberine group<fluorouracil group<5μmol/L berberine group<blank control group(P<0.01);20 μmol/L berberine group<LY294002 group<Sc79 combined with berberine group<blank control group<Sc79 group(P<0.05).3.Transwell migration assay was performed to detect the migration capacity of CNE2 cells after intervention by each group.The number of perforated cells showed blank control group>fluorouracil group>5 μmol/L berberine group>20μmol/L berberine group;Sc79 group>blank control group>Sc79 combined with berberine group>LY294002 group>20 μmol/L berberine group.4.Western blot results showed that the expression of PI3K,Akt,p-Akt,N-cadherin,and β-catenin proteins in the 20μmol/L berberine group significantly decreased compared with the control group(P<0.01),and the expression of E-cadherin proteins significantly increased compared with the control group(P<0.01).In the Sc79 combined with berberine group and LY294002 group,the expression of PI3K,Akt,p-Akt,N-cadherin,and β-catenin proteins was lower than that in the control group(P<0.05),and the expression of E-cadherin protein was higher,which was statistically significant(P<0.05).Conclusion:Berberine can inhibit the migration of nasopharyngeal carcinoma CNE2 cells,which may be related to its inhibition of PI3K/AKT pathway and regulation of the expression of downstream migration-related proteins. |
