| Ilex asprella belonging to the family Ilex,is one of representative medicinal plants in the Five Ridges of South China.The roots and rhizomes of I.asprella,which have been used to treat wind-heat cold,acute and chronic pharyngitis and sore throat,possess the effects of clearing heat and detoxifying,stimulating salvia and reducing thirst.This paper herein describes investigations of I.asprella focusing on phytochemistry,pharmacology and quality standard.Generally,this thesis is composed of the following sections:The review section summarized a phytochemical and pharmacological progress on this species,providing a foundation for studyingits pharmacologically active ingredients,quality evaluation,and further explorations.The phytochemical investigation was launched via bioassay-guided fractionation,leading to the isolation of 54 compounds including eight new lignans named asprenols A-H(1-7,13),30 compounds which were isolated from genus Ilex for the first time(8-12,14-17,21-25,2831,34,36,39,41,43,44,47-52),and 13 analogues isolated from this species for the first time(20,26,27,32,33,35,38,40,45,46,53,54).Pharmacological evaluation of these compounds were tested according to the antiinflammatory activities in RAW264.7 and BV-2 cells.Compounds 25,26,33,43 and 46 inhibited nitric oxide(NO)production in lipolysaccharide(LPS)-induced RAW 264.7 macrophage cells with IC50 values ranging from 35.71-53.91 μM.Compounds 25 and 26 showed moderate inhibitory effects against NO production in BV-2 cells with IC50 values of 57.97 and 56.34 μM,respectively.Thirty-three batches of I.asprella from different origins were analyzed by LCMS-IT-TOF to compare the chemical constituents from their roots and stems,and 13 characteristic compounds were used to evaluate the 33 batches of Ilex asprella.Results showed that the saponin and phenolic acid were the major components of I.asprella,and the cmponents of the roots and stems are pretty similar.In brief,this paper covers a phytochemical research and a preliminary vivo antiinflammatory evaluation on I.asprella.A total of 54 compounds including eight new analogues were isolated and structural detemined.Components from the roots and stems of 33 batches of I.asprella from different origins were analyzed using LC-MS,results showed that their cmponents were very similar. |
