| Mycotoxins may contaminate crops during their whole growth cycle and storage,which will cause multiple threats to human and animal health,thus affecting feed safety,food safety and even human life and property safety.From the perspective of research and analysis of antibacterial of low-concentration of Macleaya cordata extract,a series of experiments related to inhibition of the toxin production ability of mould of Macleaya cordata extract were carried out.Through UPLC-Q-TOF MS,quickly screen and sorting of a variety of mycotoxins and concealed mycotoxins were made to determine the basic conditions of mycotoxin exposure.Based on the above,this paper developed the basic research on how low-concentration of Macleaya cordata extract intervenes mold growth and toxin production ability in corn flour,analyzed the experimental performance of different concentrations of Macleaya cordata extract in inhibiting the toxin production ability of mould and demonstrated the application principle and future application value of antifungal agent of plant extract,with a view to providing a scientific reference for the intervention to toxin production ability of mould in grains produced by Chinese antifungal agents of herbal medicine.The concrete study outcome of this thesis are as below:(1)Determined the inhibitory effect of Macleaya cordata extract BE60 on Fusarium poae through paper disk method,and through the PDA agar mixture and surface bacteria coating method and the bacteriostatic method of PDA agar surrounded by pie-shaped bacteria in the middle and paper externally,the minimum inhibitory concentration(MIC)of Macleaya cordata extract BE60 against Fusarium poae was defined to be 62.5 μg/m L.(2)When different concentrations of Macleaya cordata extract BE60 were evenly sprayed in corn flour infected by Fusarium poae and the dilution ratio was1.6-16,they had a certain inhibitory effect on the growth and toxin production ability of Fusarium poae in corn flour.The effect of BE60 on the micromorphology of Fusarium poae was observed by scanning electron microscope.With the increase of BE60 concentration,the spore surface appeared abscission and crack.Macleaya extract(BE60)inhibited the colony diameter and growth rate of Aspergillus flavus.With the increase of BE60 concentration,the colony diameter gradually decreased.Through the observation under scanning electron microscope,BE60 destroyed the spore morphology and structure of Aspergillus flavus.With the increase of BE60 concentration,the conidial head alienated and gradually became smaller,the conidia became shriveled,the conidial stem cracked,and the sporulation ability decreased.(3)AFL-B1 toxin content of Aspergillus flavus cultured in PDA medium with different concentrations of BE60 for 5 days was detected.Compared with the blank control group,after adding 1 μg/m L has inhibitory effect.With the increase of concentration,the inhibitory effect gradually increases to 250 μg/m L can completely inhibit the growth and toxin production of Aspergillus flavus.It can be seen that the low concentration extract BE60 of Macleaya has an inhibitory effect on the toxin production ability of mold,but it is dosedependent. |
