Study Of Yifuzibaijiangsan And Its Decomposed Formulas On The Rats With Ulcerative Colitis

Objective:To study the effect mechanism of YFBS and its disassembly formula on the colonic mucosal repair of DSS-induced UC model rats,so as to reveal the formulation significance of YFBS and provide a scientific basis for the prevention and treatment of classic TCM prescriptions for UC.Methods:70 male SD rats were randomly divided into normal group,model group,positive group,whole prescription group,semen coicis group,Aconite group and Radix Oleifera group.Except blank group,the other 6 groups were free to drink 3% DSS solution for 7 days to prepare ulcerative colitis model.On the 8th day,the patients were given corresponding drug intervention treatment,and the subjects were sacrificed and collected after 7 days of medication.The general conditions of rats were observed,including body weight,food intake,fecal traits and fecal occluded blood,etc.DAI score,colon length,HE staining and histopathological score(HS)were recorded.The contents of IL-6,IL-10,SOD and MDA in colon tissue were detected by ELISA.The expression of PCNA,Csapase-3 and EGF in colon tissue was detected by immunohistochemistry.Western Blot was used to detect the protein expression of Occludin and ZO-1 in colon tissues.Results:(1)In the model group,rats had poor mental state,weight loss,reduced food intake,loose stools and bloody stools,DAI score was significantly increased(P<0.05),colon length was shortened,HE staining showed colonic tissue structure destruction,mucosal defect,gland separation,massive neutrophils infiltration,and HS score was increased(P<0.05).After drug intervention,the general condition of rats in the positive drug group and the traditional Chinese medicine treatment group was improved,with the decrease of DAI score and HS score.(2)Compared with model group,YFBS can effectively reduce the level of IL-6(P<0.05)and increase the level of IL-10(P<0.05).Among the three ingredients,Aconite can effectively reduce the level of IL-6(P<0.05)and increase the level of IL-10(P<0.05);The level of IL-6(P<0.05)could be effectively reduced by Dahurian Patrinia Herb,but the level of IL-10 could not be increased.Semen Coicis had no regulation effect on IL-6 and IL-10(P>0.05).(3)Compared with model group,YFBS increased SOD content(P<0.05),decreased MDA content(P<0.05),and significantly up-regulated Nrf2 protein expression(P<0.05).Among the three ingredients,Semen Coicis,Aconite and Dahurian Patrinia Herb could all increase SOD content,and the effect was similar(P>0.05).At the same time,all the three medicines could reduce the content of MDA(P<0.05),and the effect of Dahurian Patrinia Herb and Semen Coicis was similar(P>0.05),which was significantly better than that of Aconite(P<0.05).In addition,Semen Coicis and Aconite could both up-regulate the expression of Nrf2 protein(P<0.05),and Semen Coicis was better than Aconite(P<0.05),while Dahurian Patrinia Herb had no effect on the expression of Nrf2 protein(P>0.05).(4)Compared with model group,YFBS and three composition drugs could significantly up-regulate the protein expression of PCNA and EGF(P<0.05),down-regulate the protein expression of Caspase-3(P<0.05).Among the three components of drugs,Semen Coicis had the most significant effect on the up-regulation of PCNA expression and down-regulation of Caspase-3 expression(P<0.05),while the effect of Dahurian Patrinia Herb and Aconite was similar(P>0.05).As for the up-regulation of EGF protein expression,Dahurian Patrinia Herb was better than Semen Coicis and Aconite(P<0.05).(5)Compared with model group,YFBS significantly up-regulated the protein expression of Occludin and ZO-1(P<0.05).Among the drugs composed of three flavours,Semen Coicis,Aconite and Dahurian Patrinia Herb were significantly up-regulated(P<0.05),and there was no difference among the three(P>0.05).Meanwhile,Semen Coicis and Aconite both significantly up-regulated the expression of ZO-1 protein,but there was no difference between the two(P>0.05),while ZO-1 protein expression was not increased by Dahurian Patrinia Herb(P>0.05).Conclusions:(1)Both YFBS and its three-flavor components have therapeutic effects on UC model rats,which can improve the pathomorphological changes of intestinal mucosa,reduce neutrophils infiltration,and promote the repair of colon mucosa.(2)Among the three compounds of YFBS,Semen Coicis seed plays a significant role in regulating the expression of Nrf2 to reduce oxidative stress injury,promote cell proliferation,inhibit excessive apoptosis,and promote tight connection repair.Aconite has a significant effect on regulating the level of inflammatory factors and promoting the repair of tight junctions.The results showed that Dahurian Patrinia Herb had significant effects on inhibiting proinflammatory factors and regulating epidermal growth factor.(3)Through the synergistic effect of coicis,Aconite and Dahurian Patrinia Herb,YFBS plays a repairing role in colonic mucosa of rats with ulcerative colitis in terms of anti-inflammatory,anti-oxidation,promoting cell proliferation,inhibiting excessive cell apoptosis,and promoting tight junction repair.Key
