| Objective Diffuse large b-cell lymphoma(DLBCL)is a heterogeneous group of diseases in morphology,genetics,molecular biology and clinical manifestations.The standard treatment for DLBCL is R-CHOP,about 70% of elderly patients can achieve a complete response,but 30% of patients after standard treatment after recurrence.With the increasing incidence of DLBCL,the recurrence rate and mortality rate are also increasing.Although new diagnostic techniques and targeted therapeutic agents have been proposed,nearly half of newly diagnosed patients can not achieve complete remission.Therefore,it is urgent to find new potential therapeutic targets and biomarkers for the diagnosis and treatment of DLBCL.This study aims to analyzed the expression levels and significance of the apolipoprotein C1(APOC1)and apolipoprotein E(APOE)in DLBCL tissues were analyzed by bioinformatics and immunohistochemistry.Methods 1.The expression of APOC1 and APOE in pan-carcinoma was retrieved by using the TIMER 2.0 database.The TCGA database was used to analyze the expression levels of APOC1 and APOE in DLBCL,and the prognostic value of APOC1 and APOE in DLBCL was analyzed by GEPIA2 database.The APOC1 and APOE protein interaction network was constructed through the STRING database and functional annotation and pathway analysis by the Web Gestalt online analysis platform.2.Immunohistochemistry to detect the expression of APOC1 and APOE proteins in DLBCL tissues: This study collected the clinical data and pathological tissue samples of patients with first-onset DLBCL confirmed by pathological tissue biopsy in Gansu Provincial Hospital from June 2017 to June 2022,and 20 lymph node reaction proliferative tissue samples,and used immunohistochemistry to detect the expression levels of APOC1 and APOE in the two groups and analyze the relationship between the expression of the two groups and the clinicopathological data of DLBCL patients.Results The results of bioinformatics analysis showed that the expression of APOC1 and APOE in DLBCL was higher than that of normal tissues,and the difference was statistically significant(P<0.01).The enrichment analysis results showed that the genes related to APOC1 and APOE were mainly related to metabolic processes,bioregulation,multicellular biological processes,cellular component tissues,cholesterol metabolism,etc.Bioinformatics analysis showed that the overall survival of patients with high expression of APOC1 and APOE was higher than that of patients with low expression(P<0.05).The results of immunohistochemical experiments showed that the positive expression rates of APOC1 and APOE in DLBCL were 72.86% and 68.57%,and 25% and 30% in lymph node-responsive hyperplasia tissues,respectively,which were higher than those in the control group,and the differences were statistically significant(P<0.05).The expression of APOC1 and APOE in DLBCL tissues was significantly correlated with the clinical stage of patients(P<0.01),and the earlier the stage(Phase I+II),the higher the positive expression rate of APOC1 and APOE.The expression levels of APOC1 and APOE were not related to the age,gender,β2microglobulin(β2-MG),C-reactive protein(CRP),B symptoms,IPI score,etc.(P > 0.05).Conclusion1.The expression levels of APOC1 and APOE in DLBCL were higher than those of lymph node responsive hyperplastic tissues.2.The expression levels of APOC1 and APOE are related to tumor stage in DLBCL patients and may be involved in tumor development.3.The overall survival of patients with high expression of APOC1 and APOE in DLBCL is higher than that of patients with low expression,which may participate in the occurrence and development of tumors through lipid metabolism,which may provide a theoretical basis for the diagnosis and treatment of DLBCL. |
