Objective:In this study,polysaccharides from Ganoderma lucidum were extracted to study the effects of Ganoderma lucidum polysaccharide on learning and memory impairment in D-gal aging mice,explore the mechanism of its improvement on learning and memory impairment in mice,and explore its role in regulating intestinal flora in mice.Method:The extraction factors were determined by single factor experiment,and the extraction conditions were optimized by response surface test.Ganoderma lucidum polysaccharide was used as the research object to establish a mouse aging model,and it was verified that Ganoderma lucidum polysaccharide could improve learning and memory disorders in mice by detecting mouse biochemical indexes.The target of ganoderma lucidum polysaccharide to improve learning and memory disorders in mice was screened by m RNA sequencing technology.GO functional annotation analysis and KEGG pathway enrichment analysis were performed on the drug targets screened by DAVID database.Finally,the key genes and proteins of Cholinergic synapse pathway were verified by Western blot analysis.By 16 s RNA sequencing technology,Ganoderma lucidum polysaccharide can improve the intestinal flora of mice with learning and memory disorders.Results:The extraction temperature of ganoderma lucidum polysaccharide was 89℃,the ratio of solid to liquid was 1:28.9,the extraction duration was 3.2 h,the soaking time was 10.3 h,and the maximum yield reached 1.98% when the extraction times were3.Ganoderma lucidum polysaccharide can improve the antioxidant capacity,clear free radicals,reduce lipid peroxidation and oxidative stress by enhancing the activities of SOD and GSH-Px in serum of mice,increasing the contents of GLU and ACH,and reducing the contents of MDA and GABA,so as to improve the learning and memory disorders of mice.Through m RNA sequencing,644 differentially expressed genes of ganoderma lucidum polysaccharide could improve learning and memory impairment in mice.In addition,KEGG pathway analysis revealed that Cholinergic synapse pathway was an important pathway for ganoderma lucidum polysaccharide to improve learning and memory impairment in mice,and Gnb1 and Adcy2 genes in the pathway may be the key genes for Ganoderma lucidum polysaccharide to improve learning and memory impairment in mice.Other proteins in the pathway(M2,Cav-1,PI3 K,MEK,ERK,FOS,n ACh R,PKA,CREB)were also involved in the process of ganoderma polysaccharides improving learning and memory impairment in mice.Through16 s RNA sequencing,we found that ganoderma lucidum polysaccharides could affect9 kinds of intestinal flora,The Lactobacillus and Chloroalkane and chloroalkene degradation pathways may be the key microflora and related pathways for ganoderma lucidum polysaccharide to improve learning and memory impairment in mice.Conclusion:In this paper,single factor experiment and response surface design experiment were used to optimize the extraction of crude polysaccharides from Ganoderma lucidum leaves,and it was found that Ganoderma lucidum polysaccharide has antioxidant activity and can improve learning and memory disorders in mice.The mechanism is to improve learning and memory impairment in mice by regulating Gnb1 and Adcy2 genes and nine related proteins in Cholinergic synapse pathway.The improvement of learning and memory impairment in mice by Ganoderma lucidum polysaccharide may be related to its Lactobacillus in intestinal flora.
