Objective To clarify the difference of hn RNP U(heterogeneous nuclear ribonucleoprotein U)expression in normal skin and keloid tissues,to clarify the effect of hn RNP U on the biological properties of keloid fibroblasts(KFs),to investigate the relationship between hn RNP U expression levels in relation to keloid patients.Methods 1.Analyze the expression level and localization of hn RNP U in keloid tissues by immunohistochemical staining,analyze the differential expression genes of keloid tissue and normal skin tissue by using gene expression synthesis database,and synthesize the hn RNP U targeting gene in the CLIPdb database to predict the role of hn RNP U in keloid tissue.2.Isolate and culture keloid fibroblasts,and analyze the expression level of hn RNP U in KFs by Western Blot.Knockdown hn RNP U with lentivirus and plot cell proliferation curves by CCK-8assay,detect cell migration ability using cell scratch assay and Transwell migration assay.3.Analyze the relationship between the expression of hn RNP U in keloid tissue and the clinical parameters such as age,sex and course of keloid patients.Results 1.Immunohistochemical results showed that hn RNP U positive cells were more abundant and expressed in fibroblast nuclei in keloid tissues compared with normal skin(P<0.05).2.hn RNP U downstream targeting genes were enriched in various aspects such as enzyme binding,catalytic activity,RNA binding,etc.Differential gene enrichment analysis of hn RNP U targeting showed that upregulated genes were mainly concentrated in fibrous tissues,collagen biosynthesis,etc.3.Western Blot showed that hn RNP U was highly expressed in KFs(P<0.05),hn RNP U correlated with the proliferative and migratory abilities of KFs,and knockdown of hn RNP U inhibited the proliferative and migratory abilities of KFs(P<0.05).4.The expression level of hn RNP U in keloid tissue was not statistically significant compared with the clinical parameters such as age,sex and course of keloid patients.Conclusions 1.hn RNP U expression in keloid tissues was higher than that in normal skin,and it was mainly expressed in the nucleus of keloid fibroblasts.2.hn RNP U is involved in the regulation of keloid fibroblast proliferation and migration,and knocking down hn RNP U could inhibit the proliferation and migration of keloid fibroblasts.
