| Objective(s): Elsholtiza bodinieri Vaniot,as a kind of medicinal and edible plant resource in Yunnan,is rich in flavonoids,terpenoids and other chemical components.It has the functions of anti-oxidation,anti-inflammation,anti-microbial,antipyretic,analgesic,antispasmosis,and enhancing immunity.Essentialoil of Elsholtiza bodinieri Vaniot can reduce lipid in obesity model of C.elegans,but its mechanism needs other study.In this study,C3H10T1/2 lipoblast cells and PA-induced HepG2 cells were used as models to investigate the effects of essential oil of Elsholtiza bodinieri Vaniot on lipid metabolism in cells and its mechanism,so as to provide basic data for medicinal research and food development of Elsholtiza bodinieri Vaniot.Methods: A solvent-free method was used to extract essential oil from Elsholtiza bodinieri Vaniot.Lipogenesis of C3H10T1/2 cells was induced by 0.5 M IBMX,2.5m M dexamethasone,344.38 μM insulin,1 m M rosiglitazone,125 μM indomethacin,and 30.72 μM T3;The steatosis model of human hepatoma HepG2 cells was induced by 250 μmo L palmitic acid.The survival rate of cells was measured by MTT method to define the appropriate range of concentration of essential oil and Orlista.Blank control group,model control group,orlista positive control group and essential oil intervention group were set up.Oil red O staining method was used to stain the cells after intervention and the fat content was quantitatively analyzed.GPO-PAP enzyme and COD-PAP enzyme were used to detect the contents of triglyceride and cholesterol in cells treated with essential oil.The expression levels of the C/EBP-α,PPARγ,FABP4,AMPK,SIRT1,PPARα,CD36 mRNA and related proteins in the regulation of lipid metabolism were detected by quantitative reverse transcription-polymerase chain reaction gene transcription and Western Blot,respectively.To explore the effects of essential oil of Elsholtiza bodinieri Vaniot on lipid metabolism and its molecular mechanism.The data were processed by Image J,statistically analyzed by SPSS.17.0,P<0.05,indicating statistically significant differences.Results: The essential oil of Elsholtiza bodinieri Vaniot reduced the accumula tion of triglyceride and cholesterol in C3H10T1/2 lipoblasts and PA-induced He p G2 cells.The lipogenic differentiation of C3H10T1/2 cells was significantly in hibited by decreasing CCAAT enhancer binding protein α,peroxidase proliferat or activator receptor γ and fatty acid binding protein 4.These results suggest t hat the essential oil may play a regulatory role in lipid metabolism of C3H10T1/2 lipoblasts through C/EBPα/PPARγ/FABP4 signaling pathway.The essential o il can regulate the expression by increasing the silencing information1,Adenosi ne5’-monophosphate(AMP)-activated protein kinase,and Peroxide Proliferator-acti vated receptor α and its downstream fatty acid translocationenzyme protein,and significantly reduce the accumulation of lipids in HepG2 cells induced by PA.These results suggest that essential oil of Elsholtiza bodinieri Vaniot may play a role in lipid lowering of PA-induced HepG2 cells through SIRT1/AMPK sign aling pathway.In addition,the essential oil of Elsholtiza bodinieri Vaniot can al so improve the lipid toxicity induced by PA by reducing the degree of apoptos is.Conclusion(s): The essential oil of Elsholtiza bodinieri Vaniot has a certain lipid reduction effect.We suggest that the essential oil may exert inhibition on lipogenesis differentiation of C3H10T1/2 cells by regulating C/EBP-α/PPARγ/FABP4 signaling pathway.It may play a role in inhibiting PA-induced lipid metabolism of HepG2 cells by regulating SIRT1/AMPK signaling pathway.However,the specific mechanism and whether it can reduce fat content through otherways need other research. |
