| Objectives:1.To study the difference of expression levels of miR-125 b and circPVT1 in peripheral blood of patients with bone metastasis of Xuanwei lung adenocarcinoma and patients without bone metastasis of lung adenocarcinoma;2.Analyze the relationship between the expression level of miR-125 b,circPVT1 and various clinicopathological characteristics;3.To evaluate the diagnostic value of miR-125 b and circPVT1 combined with CEA and related tumor markers in the diagnosis of bone metastasis of Xuanwei lung adenocarcinoma and the relationship between them and prognosis,and analyze the relevant factors related to the risk of bone metastasis,so as to provide important theoretical basis for the early diagnosis and treatment of bone metastasis of Xuanwei lung adenocarcinoma and new intervention targets for prognosis evaluation.Methods: In this study,we collected and followed up patients with lung adenocarcinoma in Xuanwei area who were diagnosed by the Department of Pathology at Yunnan Cancer Hospital from May 2020 to September 2022.There were47 patients without bone metastasis and 34 patients with bone metastasis;Collect preoperative peripheral blood samples,detection results of relevant tumor markers,and clinicopathological data of patients;The expression of microRNA-125 b and circRNA-PVT1 in peripheral blood samples of the two groups of patients was detected using q RT-PCR.The expression of microRNA-125 b and circRNA-PVT1 in the two groups was compared using independent sample t-test and chi-square test,and the relationship between the expression of microRNA-125 b and circRNA-PVT1 in the two groups and various clinical and pathological data was analyzed;To analyze the relationship between the expression of microRNA-125 b and circRNA-PVT1 combined with CEA and related tumor markers and the diagnosis of bone metastasis in Xuanwei lung adenocarcinoma;Through follow-up and statistical analysis with SPSS25.0 and Graph Pad Prism 7.0 software,the relationship between microRNA-125 b,circRNA-PVT1,and clinicopathological data and the prognosis of patients with bone metastases from Xuanwei lung adenocarcinoma was determined;Through binary logistic regression analysis,the multivariate variables related to the risk of bone metastasis of Xuanwei lung adenocarcinoma were detected,with a test level of α= 0.05,with P<0.05 being statistically significant.Results: The relative expression of Micro RNA-125 b and circRNA-PVT1 in the blood conformed to a normal distribution;Compared with the group without bone metastasis,the expression level of microRNA-125 b in the group with bone metastasis significantly decreased,while the expression level of circRNA-PVT1 significantly increased,with significant differences(both P<0.05).After data analysis,it was found that low expression of microRNA-125 b and high expression of circRNA-PVT1 were closely related to bone metastasis,and there was a significant negative correlation between the relative expression of microRNA-125 b and circRNA-PVT1(r=-0.239,P<0.05).Using ROC(subject working curve)curve analysis,the results showed that the AUC(area under the curve)of microRNA-125 b relative expression was 0.7411± 0.07148,and the area under the curve of circRNA-PVT1 relative expression was0.8911 ± 0.04214.Both of them had high diagnostic and predictive value for bone metastasis of Xuanwei lung adenocarcinoma,among which the combined diagnostic efficacy of miR-125b+circPVT1+CEA+CYFRA21-1+NSE was the best,with an area under the curve of 0.9922 ± 0.069.The Kaplan Meier method was used to analyze the prognosis of patients.Low tumor differentiation and bone metastasis were closely related to the prognosis of patients,and the relative expression of microRNA-125 b and circRNA-PVT1 was significantly correlated with the prognosis of patients.Binary logistic regression analysis was used to detect multivariate variables related to the risk of bone metastasis in lung adenocarcinoma.miR-125 b,circPVT1,and CYFR2A1-1 were independent risk factors for bone metastasis in this analysis model.Conclusions:1.The expression of miR-125 b and circPVT1 in the blood of patients with bone metastases from Xuanwei lung adenocarcinoma was significantly different from that of patients without bone metastases.Mi R-125 b was significantly lower in the blood of patients with bone metastases from lung adenocarcinoma,while circPVT1 was significantly higher in the blood of patients with bone metastases from lung adenocarcinoma2.There is a significant negative correlation between the expression of miR-125 b and circPVT1 in the blood of Xuanwei lung adenocarcinoma patients;3.Patients with low expression of miR-125 b,high expression of circPVT1,poorly differentiated tumors,and bone metastases from Xuanwei lung adenocarcinoma have worse prognosis;4.Both miR-125 b and circPVT1 in blood have high diagnostic and predictive value for bone metastasis of Xuanwei lung adenocarcinoma,and their combination with CEA and related tumor markers has the highest diagnostic value;5.Mi R-125 b,circPVT1,and the tumor marker CYFRA21-1 in the blood are associated with the risk
