| Objective: The immune response is involved in the occurrence and development of periodontitis.Helper T cells are an important part of the immune response.As a newly discovered helper T cell subset in recent years,Th9 cells have an important impact on a variety of inflammatory diseases.However,the correlation between Th9 cells and periodontitis is still unclear.In this study,the expression levels of Th9 cell-specific differentiation markers PU.1,IRF4,SIRT1 and IL-9 in periodontal tissues(gingival tissues and gingival crevicular fluid)of periodontitis patients and periodontal healthy subjects were compared to explore the correlation between Th9 cells and periodontitis,and to lay a foundation for periodontal immunopathological mechanism.Methods: According to the new classification of periodontal disease and peri-implant disease in 2018,the subjects were divided into Peridontitis group(P group)and Periodontal health group(H group).Demographic basic data,periodontal clinical indicators and gingival crevicular fluid samples were collected(informed consent was signed,ethical review batch number : KYKQ2023MEC035);after 6 weeks of periodontal basic treatment,periodontal clinical indicators and gingival crevicular fluid samples were collected again,and the expression level of IL-9 in gingival crevicular fluid before and after treatment was detected by ELISA.The gingival tissues of patients with periodontitis and periodontal healthy patients(signed informed consent,ethical review batch number :KYKQ2023MEC035)were collected,and the expression levels of IL-9,PU.1,IRF4 and SIRT1 in gingival tissues were detected by WB.The co-localization of PU.1 and IRF4 proteins in gingival tissues was detected by immunofluorescence.Results: Compared with group H,the expression of IL-9 in gingival crevicular fluid of group P was significantly increased(P < 0.05).After periodontal basic treatment,the expression of PLI,BOP,PD and CAL and IL-9 were significantly decreased(P < 0.05).Clinical correlation analysis showed that there was a significant positive correlation between the expression level of IL-9 in gingival crevicular fluid and PLI,BOP,PD and CAL.Compared with group H,the expression of IL-9 in gingival tissue of group P was significantly increased(P < 0.05),and the expression levels of PU.1,IRF4 and SIRT1 were increased but not statistically significant(P > 0.05).Immunofluorescence staining showed that PU.1 and IRF4 were mainly distributed in the nucleus of gingival epithelial cells,and the proportion of PU.1 and IRF4 double positive cells in group P was significantly increased(P < 0.05).Conclusion: The expression of Th9 cell-specific marker factors in gingival tissue and gingival crevicular fluid of patients with periodontitis was significantly increased,which was mainly distributed in the nucleus of gingival epithelium,and there was a significant positive correlation with clinical indicators PLI,BOP,PD and CAL,suggesting that Th9 cells may have a certain regulatory effect on the occurrence and development of periodontitis,but the related mechanism needs further study. |
