Background and purposeEndometriosis(EMs)is one of the common risk factors that damage the fertility of women of childbearing age and lead to infertility,but the specific mechanism of EMs-related infertility has not yet been elucidated.In recent years,studies have found that micro-RNA(micro RNA)participates in the occurrence and development of EMs-related infertility by regulating various pathophysiological processes such as embryonic development,fertilization and implantation.Follicle fluid is an important component of follicle growth and development,and abnormal changes in its composition will ultimately affect clinical pregnancy outcomes.In this study,the expression levels of miR-135a-5p and miR-143-3p in follicular fluid were measured to explore whether there were differential expressions of the above two miRNA in follicular fluid of patients with EMs and the correlation between their expression levels and treatment indicators of assisted reproductive technology.MethodsThe follicle fluid of 25 patients with EMs who received IVF/ICSI-ET treatment in the Department of Reproductive Medicine,the First Affiliated Hospital of Harbin Medical University from September 2020 to December 2020 was collected as the EMs group,and the follicle fluid of 25 patients who were infertile due to fallopian tube factor or(and)male factor at the same time was collected as the control group.The data of age,duration of infertility,total amount of Gn,number of oocytes obtained,number of MII eggs,and high-quality embryo rate in the two groups were recorded in detail.The PCR method was used to detect the expression levels of miR-135a-5p and miR-143-3p in follicular fluid of the two groups and SPSS26.0software was used for statistical analysis.Results1.There was no statistical difference between the two groups in age,duration of infertility,BMI,b FSH,b LH,Gn day and Gn total amount(P>0.05).2.There was no significant difference in the normal fertilization rate and cleavage rate between the two groups(P>0.05),while the oocyte retrieved,MII oocyte number,high-quality embryo number,oocyte retrieved rate,high-quality embryo rate and available embryo rate in EMs group were lower than those in the control group(P<0.05).3.The expression levels of miR-135a-5p and miR-143-3p in follicular fluid of patients in the EMs group were higher and lower than those in the control group,respectively(P<0.001;P<0.001).4.The expression level of miR-135a-5p in follicle fluid was moderately and negatively correlated with the number of high-quality embryos and the high-quality embryo rate(r =-0.496,P<0.001;r=-0.409,P=0.003);There was a moderate positive correlation between the expression of miR-143-3p and the oocyte retrieval rate(r=0.326,P=0.021).Conclusions1.In this experiment,the expression levels of miR-135a-5p and miR-143-3p in follicle fluid of patients with EMs were different from those in the control group,suggesting that the abnormal expressions of miR-135a-5p and miR-143-3p in follicle fluid might be related to EMs.2.The expression level of miR-135a-5p is negatively correlated with the number of high-quality embryos and the high-quality embryo rate,while the expression level of miR-143-3p is positively correlated with the egg collection rate.These results suggested that these two miRNA might affect the oocyte and embryo development and thus the pregnancy outcome of patients with endometriosis,and provided a new idea for further improving the effect of assisted reproductive technology in patients with EMs infertility.
