for the study of TCM intervention on acute pancreatitis.Methods: The model method of acute pancreatitis was established by using urenin and sodium taurocholate to select the suitable model method.Then the hyperlipidemia model was established with high fructose combined with high fat emulsion,and the normal group and the model group were set up,and the hyperlipidemia of the rats was recorded.Two weeks after the model group was established,the hyperlipidemia in the model group was evaluated,and two doses of FTZ-L and FTZ-H,levofloxacin and fenofibrate were given by gavage every afternoon,respectively.Among them,the HLAP group was given by gavage of emulsion and high fructose water every morning.The general performance of the animals was observed during modeling and administration,and lipid levels were measured for two weeks after modeling and four weeks after administration.Four weeks after prophylactic administration,the acute pancreatitis model was established by surgical modeling with 5% sodium taurocholate.The contents of NF-κB and IL-6 and TNF-α in pancreatic tissues of rats were determined by ELISA kit.Pathomorphic changes of pancreas were analyzed by HE staining under light microscopy in part of the pancreas,and gene expression levels of COX-2 and IKK in other parts were determined by q RT-PCR.Results:1.The results of amylase test in the model rats operated with sodium taurocholic acid showed that,compared with the normal group,AMS of NIU was increased and showed more severe acute pancreatitis;Compared with the normal group,AMS was significantly increased in Rhymenin group.There was no significant difference in AMS in the saline group.2.AMS and LPS test results of HLAP model group showed that,compared with AP model group,AMS and LPS of HLAP model group were increased,showing a more serious state of acute pancreatitis;Compared with low dose of FTZ,AMS of HLAP was significantly increased.AMS was significantly reduced in the low-dose FTZ group compared with the fenofibrate group and the high-dose FTZ group.3.The results of four blood lipid tests in the HLAP model experiment showed that,compared with the AP model group,the four blood lipid tests in the HLAP model group were increased except for the decrease of HDL-C,showing the disorder of lipid metabolism,among which TC was significantly increased;HDL-C in HLAP was lower than that in fenofibrate,high-and low-dose FTZ groups.Compared with the fenofibrate group,TC and TG of rats in the low-dose FTZ group were decreased.The results of four blood lipid tests of rats in the HLAP model experiment showed that,compared with the AP model group,the levels of four blood lipids in the HLAP model group were increased except for HDL-C,showing lipid metabolism disorders,among which TC was significantly increased.Compared with the fenofibrate group,the HDL-C of HLAP was decreased in both the high-dose and low-dose FTZ groups.Compared with the fenofibrate group,the TC and TG of the low-dose FTZ group were decreased.LDL-C was significantly lower in the high-dose FTZ group than in the fenofibrate group.Conclusion: Hyperlipidemia can aggravate the inflammatory state of pancreatitis,and the effects may be bi-directional.FTZ may play a role in the prevention and treatment of hyperlipidemic pancreatitis through the NF-κB inflammatory pathway... |
