Objectives: Clarify the levels of changes in rat epidermal cell proliferation and autophagy induced by burn serum intervention,and investigate whether Tanggulat burn solution yak Achilles tendon collagen composite sponge can participate in regulating the biological behavior of rat epidermal cell proliferation and autophagy induced by burn serum intervention.Methods: Set up Simulated burn model and burn model of laboratory rats.Male SD rats were grouped into 3 groups :normal serum group,simulated burn group and burn group.After the sample blood was taken through the abdominal aorta by using Venous blood collection vessels,Label for backup and centrifuged.The rat epidermal cell resuscitation was passaged to 3rd generation and placed in the Petri dish for Synchronizedtreatment.The epidermal cells were intervened with normal serum,simulated burn serum,burn serum,burn serum + Vaseline gauze,burn serum+ Tanggulat burn liquid collagen complex sponge for 24 hours.The experimental groups were as follows: normal serum group(blank control group),simulated burn serum group,burn serum group,burn serum +Vaseline gauze group,burn serum + Tanggulat burn liquid yak Achilles tendon collagen complex sponge group(hereinafter referred to as complex sponge group).The cell proliferation rate was detected by CCK-8 method,PCNA expression levels were detected by Elisa method,and LC3 and Beclin-1 expression levels were detected by Western Blot method.Results: 1.The proliferation rate and PCNA protein concentration of rat epidermal cells in each group could be detected under different intervention conditions,and the following conclusions could be drawn:(1)The cell proliferation rate and PCNA protein concentration of different treatment groups(burn serum group,burn serum group + Vaseline gauze group,burn serum + composite sponge group)were lower than those in normal serum group;(2)The cell proliferation rate and PCNA protein concentration of different treatment groups(burn serum group,burn serum group + Vaseline gauze group,burn serum + composite sponge group)were lower than those in simulated burn serum group;(3)The cell proliferation rate and PCNA protein concentration of burn serum + composite sponge group were higher than those in burn serum group;(4)The proliferation rate and PCNA protein concentration of burn serum + compound sponge cells were higher than those of burn serum + Vaseline gauze group.The differences between the above groups were statistically significant(P<0.01).2.The expression of Beclin-1 and LC3 protein in rat epidermal cells in each group under different intervention conditions could be concluded respectively:(1)The expression of Beclin-1 and LC3 protein in different treatment groups(burn serum group,burn serum group + Vaseline gauze group,burn serum +composite sponge group)was higher than that in the normal serum group;(2)The expression of Beclin-1 and LC3 proteins in different treatment groups(burn serum group,burn serum group + Vaseline gauze group,burn serum + composite sponge group)was higher than that in simulated burn serum group;(3)The expression of Beclin-1 and LC3 proteins in the burn serum + composite sponge group was higher than that in the burn serum group;(4)The expression of Beclin-1 and LC3 proteins in the burn serum +complex sponge group was higher than those in the burn serum + Vaseline gauze group.The differences between the above groups were statistically significant(P<0.001).Conclusions:(1)In the early stage of burn serum intervention,the proliferation rate and PCNA expression of epidermal cells in rats decreased,while the expression of autophagy related factors LC3 and Beclin-1increased;(2)Tanggulat burn fluid and yak Achilles tendon collagen composite sponge can regulate the proliferation rate of epidermal cells,expression levels of PCNA,LC3,and Beclin-1 proteins after burn serum intervention.
