Background and purposeBladder cancer(BLCA)is one of the urinary system tumors with high incidence rate.Although routine clinical diagnosis and treatment methods have improved the survival rate of patients,but the prognosis of patients is still poor due to tumor recurrence and metastasis.Lnc RNA plays an important role in regulating tumorigenesis and cancer immunity,and can be used as a novel marker to predict patient prognosis and immune response.Based on bioinformatics analysis,this study constructs a BLCA prognosis model and screens for targeted molecular drugs in BLCA.MethodsRNA-seq data and the corresponding clinical data were downloaded and organized from the database of The Cancer Genome Atlas(TCGA),and the patient cohorts were assigned into train cohort and test cohort.The immune related lnc RNAs(IR-lnc RNAs)were extracted based on the method of weighted gene coexpression network analysis(WGCNA).The model was constructed by LASSO regression analysis,and tumor correlation of prognostic gene set was analyzed by National Center for Biotechnology Information(NCBI)database and pan-cancer data.Also,the prognostic accuracy of the model was evaluated according to Kaplan-Meier(KM)analysis and ROC curve.Afterwards the samples were classified into low-risk groups and high-risk groups according to risk scores,and then high-risk genes were identified by differentially expressed genes(DEGs).Finally,the CMap was used to screen for small molecule drugs that could suppress the expression of high-risk genes,so as to reduce the risk of BLCA patients and improve the survival rate of patients.ResultsBased on 414 tumor samples and 19 normal samples in the TCGA database,a total of 1,342 IRGs and 409 clinical sample data were pretreated,and 927 survival data were obtained through WGCNA.The results of Pearson correlation analysis and univariate Cox regression analysis indicated that 74 IR-lnc RNAs linked to prognosis were screened out(|R|>0.4,P<0.05).Finally,12 IR-lnc RNAs and 21 IRGs were performed by LASSO regression analysis.According to the follow-up analysis,the prediction model can predict the survival and prognosis of patients in high-risk and low-risk groups.In addition,this study screen 11 potential targeted drugs of BLCA.ConclusionsThis study constructed a prognosis model based on IRGs and IR-lnc RNAs for BLCA patients,also screened 11 potential targeted molecular drugs,and provided new ideas for the precise treatment of BLCA patients.
