| ObjectiveTo observe whether loss of controllable stress can induce learning and memory im Pairment behavior,and to ex Plore the effect and mechanism of acu Puncture on learning and memory im Pairment behavior.As the Glu system is closely related to cognitive function,the intervention effect of electroacu Puncture was ex Plored by detecting the ex Pression levels of VGLUT1 rece Ptor,VGLUT2 rece Ptor and Syna Psin I Protein in the Prefrontal lobe and thalamus of mice after loss of controllable stress,and the chemical geneurons in the mPFC area could im Prove cognitive im Pairment.MethodsSPF healthy adult male C57BL/6J mice were randomly divided into control grou P,LOC model grou P,Yoked model grou P and LOC model Plus electroacu Puncture grou P.The Skinner box 7-day LOC model paradigm was used to make the model,and then electroacu Puncture in the ear nail area was Performed for 5 days.After that,the cognitive function of the mice was evaluated by o Pen field,new object and new location recognition,social interaction and Morris water maze.Then Golgi staining was used to observe the structural changes of mPFC neurons,immunohistochemical staining was used to observe the ex Pression of C-fos in the brain area of mPFC,Western blot was used to detect the ex Pression of VGLUT1,VGLUT2 and SYN-1,and the changes of corres Ponding indexes after acu Puncture.Results1.In LOC grou P,the retrieval of recognition memory had no obvious effect on the acquisition and consolidation of recognition memory,acupuncture could improve the acquisition and consolidation of memory,the expression levels of VGLUT1,VGLUT2 and SYN-1 in mp FC and MD brain decreased,and the density of dendritic spines of mp FC neurons decreased significantly.2.Activation of Glu neurons in mp FC brain region of excitatory virus mice can improve cognitive and learning and memory impairment.For mice injected with inhibitory virus,electroacupuncture can not reverse the recovery of learning and memory impairment.Conclusions1.The cognitive function and learning and memory impairment were induced in mice after cognitive failure.2.EA can effectively improve the cognitive function and the impairment of learning and memory after cognitive failure,and its mechanism may be realized by regulating the protein expression levels of VGLUT1,VGLUT2 and SYN-1 in ventral prefrontal lobe and thalamus.3.Activation of Glu neurons in mp FC can improve learning and memory impairment after cognitive failure,while inhibition of Glu neurons in mp FC aggravates learning and memory impairment,which can not be reversed by electroacupuncture.It is further explained that electroacupuncture is mainly used to treat learning and memory impairment after cognitive failure by regulating the activity level of Glu neurons in mp FC. |
