| Objective(s): This study was to investigate the expression of IL-4,IL-5and Il-13,CD44 and osteopontin(OPN)secreted by Th2 cells in allergic rhinitis.Methods: Mouse age 6 weeks old and female SPF-level BALB/C mice were selected,and mice in the experimental group were injected intraperitoneally with 200μl of PBS containing 200μg of ovalbumin(OVA)and2mg of aluminum hydroxide on days 0,7 and 14 of the experiment,and 10ug/ul OVA was used for nasal excitation for 7 consecutive days from 21 days,and10 ul was injected into each nostril each time to establish an animal model of allergic rhinitis.The control group was given the same dose of PBS each time according to the above steps.After the last OVA challenge,the two groups of animals were counted for 15 minutes of nose scratching and sneezing,and they were sacrificed after 24 hours,and relevant specimens were collected,and changes in serum IL-4,IL-5,IL-13 and OVA-SIg E were detected by enzyme-linked immunosorbent(ELISA)and Quantitative Real-time PCR,and CD44-m RNA and OPN-m RNA changes in the nasal mucosa of mice.Nasal pathological HE staining analyzed the expression of eosinophils,and nasal tissue immunohistochemistry analyzed the positive rate of OPN expression.Result(s): The number of nasal scratches and sneezing in mouse models of allergic rhinitis successfully induced by OVA increased,which was manifested as a typical "baldness" phenomenon,and this phenomenon began to gradually become obvious before the third intraperitoneal injection,and the expression of serum Th2 cytokines IL-4,IL-5,IL-13 and OVA-SIg E was increased in mice in the AR group compared with the healthy control group.Increased expression of CD44 m RNA and OPN m RNA in the nasal mucosa,and increased expression of eosinophils in the nasal mucosa;OPN expression in nasal tissues increased,and the positive rate increased.Conclusion(s):The mouse model of allergic rhinitis successfully induced by OVA showed a typical "baldness" phenomenon,that is,the symptomatology manifestations(including nose scratching and sneezing)of AR group are obvious.The increase in Th2 cytokines,CD44 and OPN expression in AR mice suggests that OPN/CD44 may be involved in Th2-mediated nasal allergy. |
