Purpose:To explore the expression and significance of G protein-coupled estrogen receptor(GPER)and c-fos in endometrial polyps,tissues adjacent to polyps and normal endometrial tissues,and to explore the possible role of GPER and c-fos in the pathogenesis of endometrial polyps,so as to provide help for clinical diagnosis and treatment of polyps,prevention of polyp malignant transformation and reduction of polyp recurrence.Method:Specimens were taken from patients who underwent hysteroscopic surgery in our hospital from January 2019 to June 2020.Endometrial polyp tissues of 30 patients with endometrial polyps(labeled as EP group),adjacent polyp endometrial tissue of the same patient(labeled as adjacent polyp group)and 24 normal endometrial tissues(labeled as control group)were taken respectively.The two-step method of immunohistochemistry(Envision detection system)was used.To detect the expression levels of GPER and c-fos in the glandular and interstitial components of endometrial polyps,polyp-adjacent endometrial tissues and normal endometrial tissues,and to detect the expression levels of GPER and c-fos in the glandular and interstitial components of endometrial polyps and normal endometrial tissues in the proliferative and secretory stages,and conduct statistical analysis.Result:1.GPER is mainly expressed in the cytoplasm of uterine glandular components.C-fos is mainly expressed in the nuclei of the glandular components of the uterus.2.The expression levels of GPER in the glandular components of EP and parapolypoid tissues were significantly higher than those in the glandular components of normal endometrial tissue,and there was a statistical difference between the two(P = 0.000).There was no significant difference in the expression of GPER in the glandular components of EP and parapolypoid tissues(P=0.280).The expression levels of GPER in the interstitial components of EP and parapolyposis tissues were significantly higher than those of normal endometrial tissues in the interstitial component,and there was statistical difference between the two(P= 0.000).There was no significant difference in the expression of GPER between EP and the interstitial component of the parapolypoid tissue(P=0.650).3.The expression of C-fos in EP gland components was significantly higher than that in tissues adjacent to polyps and normal endometrium,with statistical difference(P= 0.000).There was no significant difference in the expression of c-fos in the glandular components of parapolyp tissue and normal endometrial tissue(P=0.361).There was a significant difference in the expression of c-fos between EP and normal endometrium(P= 0.010).There was no significant difference in the expression of c-fos between EP and the interstitial components of the tissues adjacent to polyps(P=0.442).There was no significant difference in the expression of c-fos in the stroma between parapolyp tissue and normal endometrial tissue(P=0.059).4.The expressions of GPER and c-fos in the glandular and interstitial components of EP in the proliferative and secretory stages were not statistically significant(P>0.05).5.The expression level of GPER in the glandular components of normal endometrial tissue in the proliferative phase was significantly higher than that in the secretory phase(P<0.05).There was no significant difference in the expression of GPER between the proliferative phase and the secretory phase in the interstitial components of normal endometrial tissues(P>0.05).The expression levels of c-fos in glandular and interstitial components of normal endometrium in the proliferative and secretory phases were not statistically significant(P>0.05).6.In the EP group,the expression of GPER positively correlated with that of c-fos(r=0.469,P=0.009).Conclusion:1.The expression of GPER and c-fos in endometrial polyps is significantly higher than that in normal endometrial tissues,suggesting that the high expression of gper and c-fos may be related to the occurrence and development of endometrial polyps.2.In endometrial polyps,the expression of GPER is positively correlated with the expression of c-fos protein.3.The expression of GPER in normal endometrial glands may be related to menstrual cycle.
