| Circular RNA(circRNA)shows great potential in the diagnosis and treatment of diseases due to its stable circular structure.Mounting evidence suggests that circular RNAs play important roles in the development and progression of gliomas.However,its functions and underline mechanisms is still unveiled.Through circRNA array analysis based on online database,we found that circXPO1(hsa_circ_102737)was significantly upregulated in glioblastomas(GBM),and following q PCR analysis verified that circXPO1 expression was increased in both GBM tissues and cell lines.Functional studies demonstrated that knockdown of circXPO1 in GBM cells drastically suppressed cell proliferation and migration.Conversely,overexpression of circXPO1 further promoted the malignancy of GBM cells.In line with these findings,circXPO1 inhibition effectively prevented gliomagenesis in the in-situ transplantation model of nude mice.Studies have shown that circRNA mainly acts through(1)miRNA sponge;(2)RNA binding protein(RBP)regulates gene transcription or protein interaction;(3)Encode short peptides to play corresponding biological functions.Both our bioinformatic analyses and dual-luciferase reporter assays indicated that circXPO1 directly sponged miR-7-5p which acted as a tumor suppressor by negative regulation of RAF1.In conclusion,our studies suggest that the circXPO1/miR-7-5p/RAF1 axis plays key roles in GBM and may be a potential therapeutic target for GBM treatments. |
