Objective:The study analyzed the relationship between the expression level of MTHFD1 L and the clinicopathological characteristics of patients with hypopharyngeal squamous cell carcinoma by detecting the expression level of MTHFD1 L in hypopharyngeal squamous cell carcinoma tissues and their paraneoplastic tissues with immunohistochemistry and Western blotting,and provided a theoretical basis for further research on the development mechanism of hypopharyngeal carcinoma and new therapeutic targets.Methods:GEPIA database was used to analyze whether MTHFD1 L expression was different between head and neck squamous cell carcinoma tissues and normal head and neck tissues,and the relationship between MTHFD1 L expression and the prognosis of patients with head and neck squamous cell carcinoma was analyzed.The clinicopathological information of 36 patients with hypopharyngeal squamous cell carcinoma included in the study was collected,and the expression of MTHFD1 L in cancer tissues and adjacent normal tissues was detected by immunohistochemistry.The clinicopathological characteristics of the patients were analyzed.Kaplan-Meier survival curve analysis and COX univariate and multivariate regression tests were used to explore the risk factors affecting the prognosis of patients with hypopharyngeal carcinoma and the clinical prognostic value of MTHFD1 L.Western blotting was used to detect the expression of MTHFD1 L in 7 pairs of hypopharyngeal squamous cell carcinoma tissues and adjacent normal tissues.Results:1.The expression of MTHFD1 L in head and neck squamous cell carcinoma tissues is higher than that in normal head and neck tissues,and there is a difference.Patients with low expression of head and neck squamous cell carcinoma and high expression of MTHFD1 L have a better prognosis.2.Compared with normal tissues adjacent to cancer,MTHFD1 L was highly expressed in hypopharyngeal cancer tissues,which was statistically significant.3.There was no statistically significant difference between MTHFD1 L expression level and clinicopathological characteristics such as patient’s gender,age,whether they smoked and drank alcohol,pathological grade,AJCC TNM stage of tumor,and whether they needed adjuvant therapy after surgery.4.MTHFD1 L expression and postoperative adjuvant chemoradiotherapy are independent risk factors for the prognosis of hypopharyngeal carcinoma.5.The overall survival time of patients with HSCC in MTHFD1 L high expression group was significantly shorter than that in MTHFD1 L low expression group.Conclusion:1.MTHFD1 L expression was significantly enhanced in hypopharyngeal squamous cell carcinoma tissues.2.MTHFD1 L expression is an independent risk factor for overall survival in patients with hypopharyngeal squamous cell carcinoma.Compared with the low expression group,MTHFD1 L high expression group has a poor prognosis of hypopharyngeal squamous cell carcinoma,suggesting that MTHFD1 L may be a potential therapeutic target for hypopharyngeal squamous cell carcinoma in the future.
