Background and Aim:Circulating exosomal long non-coding RNA(lncRNA)plays an important role in many diseases However,little is known about their role in coronary artery disease(CAD)The aim of this study was to investigate the relationship between circulating exosomal lncRNA and coronary artery disease and to identify abnormally expressed lncRNA as a biomarker for the diagnosis of coronary artery disease.Methods:1.Study subjects: This study included 168 patients who underwent coronary angiography in Shenzhen Longgang Central Hospital and diagnosed coronary artery disease(98 cases);And coronary angiography showed normal coronary arteries in 70 subjects Clinical data have been recorded in detail in all studies Three CAD patients and three controls were selected by using a random number table and then used for gene chip analysis in the screening stage.2.Reverse transcriptase polymerase chain reaction(q RT-PCR): 20 CAD patients and20 control patients were also selected using a random number table to extract RNAfrom plasma exosomal precipitates using commercial kits to measure the target LncRNA using real-time quantitative PCR(q RT-PCR).The target LncRNA was relatively quantitatively processed to determine its relative expression in different samples.3.The diagnostic efficacy of circulating exosomes lncRNA-ST20-AS1 was analyzed by constructing the area under the receiver operating character(ROC)curve.4.Collect clinical data of patients and analyze the relationship between plasma circulating exosomes lncRNA-ST20-AS1 and clinical features by chi-square test.5.Statistical analysis: The statistical software IBM SPSS 22.0 and Graph Pad Prism were used to analyze the data Measurement data are expressed by mean ± standard deviation,and comparison is carried out by t test Chi-square test was used to compare the counting data P < 0.05 was statistically significant.Results:1.Baseline data of the two groups: There was no significant difference in age,sex and body mass index between the two groups(p > 0.05)Compared with the control group,patients with coronary artery disease had lower high density lipoprotein level(P=0.003)and higher low density lipoprotein level(P=0.043).2.By using Affymetrix microarray,48 up-regulated and 60 down-regulated LNC RNA were screened out from the exosomes of CAD patients and normal controls.3.The down-regulation of LncRNA-ST20-AS1 expression in CAD patients is expected to be a new biomarker of CAD4.The area under the curve of LncRNA-ST20-AS1 for the diagnosis of coronary artery disease was 0.643(95% CI = 0.607-0.801 P% < 0.001).5.Correlation analysis between LncRNA-ST20-AS1 and clinical markers: The level of ST20-AS1 encapsulated in plasma exosomes was positively correlated with lipoprotein a and low density lipoprotein,but there was no significant correlation with other clinical markers.Conclusions:1.Circulating exosomes LncRNA-ST20-AS1 may be a potential molecular biological marker for the diagnosis of coronary artery disease.2.Circulating exosomes LncRNA-ST20-AS1 may play a role in the diagnosis of coronary artery disease.
