ObjectiveTo analyze the influence of PEAK1 on the migration and invasion of NSCLC.Next to compare the expression of PEAK1 in distant metastases of NSCLC and the primary lesions,and analyze the clinicopathological features of PEAK1 expression and distant metastases of NSCLC patients.Further,to explore the influence and mechanism of its changes on distant metastasis and CTCs.Methods1.The expression of PEAK1 in H1299 cell line was increased by lentivirus infection,and its expression was verified by q RT-PCR and Western blot.The effect of PEAK1 overexpression on the migration and invasion of NSCLC was explored by scratch test and Transwell invasion test.2.Cases of distant metastasis of NSCLC diagnosed in the 940 th Hospital of the Joint Logistics Support Force of the Chinese People’s Liberation Army from 2012 to 2022 were collected,including 7 cases of primary lesions and distant metastasis,and 56 cases of distant metastasis.The cases were stained by immunohistochemistry,and the expression level of PEAK1 in the primary and distant metastases of NSCLC and its correlation with clinicopathologic features were analyzed by χ~2 test.3.GeneMANIA database was used to search the genes related to PEAK1,and to explore the molecular mechanism of PEAK1 in distant metastasis of NSCLC.Results1.The migration and invasion ability of the PEAK1 overexpression group was stronger than that of the empty vector group,and the difference was statistically significant(P < 0.05).2.PEAK1 expression was localized in the nucleus,and the strong positive expression rate was 92.9%(52/56)in distant metastatic tissues of NSCLC,and the weak positive expression rate was 7.1%(4/56).The strong positive expression rate was 14.3%(1/7)in primary tissues of NSCLC,and the weak positive expression rate was 85.7%(6/7).The expression intensity of PEAK1 in metastatic lesion was significantly higher than that in primary lesion,and the difference was statistically significant(P < 0.05).And it was not associated with the clinicopathologic characteristics of the patient,such as gender,age,Ki67 proliferation index,pathological classification and metastatic site.3.According to GeneMANIA database analysis,PEAK1 had a certain correlation with EGFR and ZEB1Conclusion1.In non-small cell lung cancer cells,overexpression of PEAK1 can promote the migration and invasion ability of non-small cell lung cancer cells.2.In non-small cell lung cancer tissues,the expression of PEAK1 in distant metastases is significantly higher than that in primary tumors.And its change is related to the occurrence of distant metastasis of non-small cell lung cancer.3.PEAK1 is correlated with EGFR and ZEB1,which might provide a new theoretical basis for the diagnosis and treatment of non-small cell lung cancer. |
