Objective:Head and neck cancer is the sixth most common malignancy in the world.Of these,90%of head and neck cancers are squamous cell carcinomas.The difficulty in early diagnosis of head and neck squamous cell carcinoma,the tendency to infiltrate and metastasise in later stages,and the low level of patient satisfaction with radiotherapy have all contributed to the lack of significant improvement in the overall 5-year survival rate of patients with head and neck squamous cell carcinoma.Long non-coding RNA(LncRNA)has been reported to influence the development of many cancers.Therefore,this study aims to analyse the expression of LncRNA MNX1-AS1 in head and neck squamous carcinoma tissues in combination with the clinicopathological parameters of head and neck squamous carcinoma patients,and further clarify the role of LncRNA MNX1-AS1 in head and neck squamous carcinoma cell lines through cell function assays,so as to provide new targets for the diagnosis,treatment and prognosis of head and neck squamous carcinoma.Methods:First,the head and neck tumour transcriptome sequencing data from TCGA(The cancer genome atlas)database were used to analyse the expression of LncRNA MNX1-AS1 in head and neck squamous carcinoma and normal control tissues,and the relationship with clinicopathological parameters.The expression levels of LncRNA MNX1-AS1 in human embryonic lung diploid cells 2BS and human immortalized epidermal cells Ha Ca T and head and neck squamous carcinoma cell lines FD-LSC-1,CAL-27 and Detroit 562 were examined using real-time quantitative PCR(q PCR).Secondly,the expression of LncRNA MNX1-AS1 was silenced using RNAi(RNA interference)technology,and its effects on cell biological functions were examined by CCK-8,clone formation,Transwell invasion and migration assays and flow cytometry.Results:The expression level of LncRNA MNX1-AS1 in head and neck squamous carcinoma tissues was significantly higher than that in normal control tissues(head and neck squamous carcinoma tissues;normal control tissues = 0.323;0.088),and the difference was statistically significant(P<0.05);its expression level was positively correlated with the pathological grade of head and neck squamous carcinoma patients(P<0.05).Survival analysis revealed that the overall survival rate of patients in the high LncRNA MNX1-AS1 expression group was significantly lower than that of patients in the low expression group(P<0.05),with a median survival time of 42.97 months and 61.27 months respectively.Clinical correlation analysis revealed that the expression level was higher in the group of patients with smoking history than in the group of patients without smoking history(P<0.05);the expression level was higher in the group of patients with mid-to late-stage clinical disease than in the group of patients with early-to mid-stage clinical disease(P<0.05).Knockdown of LncRNA MNX1-AS1 significantly inhibited the proliferation viability,clone formation,migration and invasion ability of head and neck squamous carcinoma cell lines(P<0.05).Conclusion:Overexpression of LncRNA MNX1-AS1 is closely related to the malignant development of head and neck squamous carcinoma and can promote the proliferation viability,migration and invasion ability of head and neck squamous carcinoma cell lines,which is a potentially important molecular marker in the treatment of head and neck squamous carcinoma.
