Objective:To investigate the role of Ubiquitously expressed transcript(UXT)in the proliferation,migration and invasion of Hepatocellular carcinoma(HCC).Methods:Firstly,bioinformatics analysis was used to verify whether there was a difference in the expression level of UXT in HCC tissues and normal liver tissues.Then the HCC cell lines with higher UXT expression levels were screened by Western Blot,and the knockdown cell model was constructed by small interfering RNA(si RNA).Finally,CCK-8 assay,cell scratch assay and Transwell assay were performed on the UXT knockdown group and the blank plasmid group to verify the effects of UXT on the proliferation,migration and invasion of HCC cells.Results:The results of bioinformatics analysis showed that the expression level of UXT in HCC tissues was significantly higher than that in normal tissues and matched adjacent tissues.The expression level of UXT in different HCC cell lines was detected by Western Blot,and it was found that the expression level of UXT was higher in Bel-7404 cell line.Then it was transfected with si RNA to construct the knockdown cell model,and the results showed that the expression level of UXT protein in si-UXT#3 knockdown group was significantly reduced.Finally,CCK-8 assay,cell scratch assay and Transwell assay showed that knockdown of UXT expression inhibited the proliferation,migration and invasion of HCC cells.Conclusion:UXT is highly expressed in HCC tissues,which is related to the proliferation,migration and invasion of HCC cells. |
