Objective:To investigate the differential expression of unconventional profolin RPB5 interbody 1(URI1)in hepatocellular carcinoma(HCC),its effect on the basal phenotype of HCC cells,and its potential prognostic value.Methods:Bioinformatics analysis was performed using cancer genome map,genotype-tissue expression,HCCDB,TISIDB,tumor immune resources.Western blotting and immunohistochemical staining were used to determine the relative levels of URI1 in hepatocellular carcinoma cell lines as well as in HCC tissues and adjacent peritumor tissues.Cell counting kit CCK-8,migration assay and scratch assay were performed to verify URI1 biological function.Results:Bioinformatics analysis showed that URI1 expression was increased in HCC.Knockdown URI1 inhibited the proliferation and migration of hepatoma cells.URI1 was significantly correlated with clinicopathologic features.Survival analysis showed that URI1 had significant prognostic value in HCC patients.The histogram and nomogram show the accuracy of URI1 as a prediction model.At the same time,enrichment analysis showed that URI1 was associated with stimulation of neural tissue interaction and Wnt signaling pathway.Immunoinfiltration analysis showed that URI1 was correlated with tumor purity,eosinophilic cells,macrophages and B cells.ROC curve showed that URI1 had good diagnostic value.Conclusions:The silencing of URI1 can inhibit the proliferation and migration of HCC cells,and URI1 can be used as a prognostic marker for HCC.These findings provide a new target for the treatment of HCC.
